甘薯及其提取物辅助改善小鼠学习记忆功能实验研究

【摘要】 研究甘薯的冻干粉及其提取物对32周龄KM雌性小鼠学习记忆能力的影响和作用。所有小鼠被分为空白对照组、甘薯冻干粉组、甘薯乙醇提取物组、甘薯精制物组,当受试物浓度分别以甘薯冻干粉750 mg/kg、甘薯乙醇提取物250 mg/kg、甘薯精制物250 mg/kg剂量连续灌胃6周后采用电击跳台和水迷宫实验测试各组小鼠学习记忆能力及各组血清、脑中的谷胱甘肽—过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、乙酰胆碱转移酶(ChAT)、乙酰胆碱脂酶(TChE)、5-羟色胺(5-HT)、去甲肾上腺素(NE)、丙二醛(MDA)和单胺氧化酶(MAO)水平。结果显示实验组与空白对照组相比,均能提高小鼠学习记忆能力和GSH-Px、SOD、ChAT、5-HT、NE含量并降低MDA、TChE、MAO含量。说明这3种物质可能通过抑制抗氧化损伤、降低TchE,提高中枢胆碱能神经功能以及抑制MAO,提高单胺类神经递质含量,从而改善小鼠的学习记忆功能。

【Abstract】 To study the influence of sweet potato freeze-dried powder and its extracts on learning and memory ability in 32 weeks old KM male mice,all mice were divided into 4 groups: control group,sweet potato freeze-dried powder group,sweet potato alcohol extracts group,and sweet potato purified extract group.After being given sweet potato freeze-dried powder 750mg/kg,sweet potato alcohol extract 250mg/kg and sweet potato purified extract 250mg/kg for six weeks respectively,each group was tested for learning and memory ability and GSH peroxidase(GSH-Px),superoxide dismutase(SOD),acetylcholine transferase(ChAT),acetylchoinesterase(TChE),5-hydroxytryptamine(5-HT),norepinephrine(NE),malonaldehyde(MDA)and monoamine oxidase(MAO) in cerebrum and serum by stand jumping and Morris water maze test.The results indicated that the experimental groups were significantly improved in the learning and memory ability,the contents of GSH-Px,SOD,ChAT,5-HT,NE were increased and the contents of MAO,TChE,MDA were decreased than control group.It showed the above 3 materirals can improve the learning and memory ability in mice by inhibiting oxidative damage,eliminating oxygen free radicals,increasing cholinergic nerve system function and inhibiting monoamine oxidate to increase the contents of monoamines nerve transmittance etc.
