【摘要】 目的:建立细辛饮片中马兜铃酸A的痕量测定方法。方法:采用UPLC-UV法,色谱柱为Acquity UPLCTM HSS T3色谱柱(2.1mm×50mm,1.8μm);流动相为乙腈(A)-0.1%甲酸(B),梯度洗脱(0min,30%A;5min,60%A;7min,100%A),流速0.5mL.min-1;检测波长251nm;柱温:40℃。结果:马兜铃酸A浓度在18.12～1450ng.mL-1范围内线性关系良好(r=0.9999),最低检测限为10.17ng.mL-1。结论:细辛饮片中马兜铃酸A的含量极低;本法专属性强,结果稳定,方法的回收率良好,可用于中药饮片细辛中的马兜铃酸A痕量检查。更多还原

【Abstract】 Objective:To develop an UPLC-UV method for the determination of trace amount aristolochic acid A in sliced Asarum.Methods:An Acquity UPLCTM HSS T3 column(2.1 mm×50 mm,1.8 μm)was adopted;The mobile phase consisted of acetonitrile(A)-0.1% formic acid(B)with gradient elution(0 min,30% A;5 min,60% A;7 min,100% A)at a flow rate of 0.5 mL· min-1;The detection wavelength was 251 nm,and column temperature was 40 ℃.Results:The calibration curve was linear in the range of 18.12-1450 ng·mL-1 of aristolochic acid A with correlation coefficient of 0.9999 and the LOD was 10.17 ng·mL-1.Conclusion:The content of aristolochic acid A in sliced Asarum is very low.The method is accurate and reproducible,and can be applied in the determination of trace amount aristolochic acid A in sliced Asarum.更多还原
