【摘要】 目的:探讨EpicsXL型流式细胞仪进行念珠菌快速药敏试验的可行性。方法:应用EpicsXL型流式细胞仪检测185株念珠菌对二性霉素B、氟康唑、伊曲康唑、酮康唑、5-FC等药物的耐药性,与M27-A标准化药敏试验结果进行比较。结果:EpicsXL型流式细胞仪可以很好地鉴别死、活念珠菌。2种药敏结果相符,分别为:二性霉素90.8%、氟康唑94.6%、伊曲康唑91.4%、酮康唑87.0%、5-FC89.2%。以M27-A药敏结果为评价标准,流式细胞术进行5种药物敏感性试验的灵敏度、特异性分别为二性霉素B:83.8%(57/68)、94.9%(111/117);氟康唑:92.5%(74/80)、96.2%(101/105);伊曲康唑:86.5%(45/52)、93.2%(124/133);酮康唑:90.4%(85/94)、83.5%(76/91);5-FC:77.5%(31/40)、92.4%(134/145)。结论:EpicsXL型流式细胞仪进行念珠菌快速药敏试验与常规药敏试验结果具有较好的一致性,且快速、敏感。更多还原

【Abstract】 Objective To develop a rapid method for antifungal susceptibility testing by EpicsXL Flow Cytometry(FCM).Methods The resistance of 185 candidas to five antimicrobials was detected by flow cytometry,and the results were compared with the M27-A method.Results Concordance rates of the susceptibility to Amphotericin B,Fluconazole、 Itraconazole,Ketoconazole and 5-FC in 185 candidas detected by EpicsXL and M27-A were 90.8%,94.6%,91.4%,87.0%,89.2% respectively.If the results of the M27-A method was taken as the golden standard,the sensitivity and specificity for Amphotericin B detected by EpicsXL ware 83.8%(57/68)and 94.9%(111/117),for Fluconazole were 92.5%(74/80)and 96.2%(101/105),for Itraconazole were 86.5%(45/52)and 93.2%(124/133),for Ketoconazole were 90.4%(85/94)and 83.5%(76/91),and for 5-FC were 77.5%(31/40)and 92.4%(134/145).Conclusion EpicsXL,being simple and sensitive,can be used as a rapid method for candida species susceptibility testing.更多还原
