贵州12个辣椒杂交种SSR标记指纹图谱的构建与分析

【摘要】 从辣椒基因组的75个SSR标记中筛选出15个多态性较好的标记,用于构建12个辣椒杂交种的数字化指纹图谱。在构建指纹图谱的基础上,用每个标记的指纹数据分别作聚类分析,结果表明:标记Hpms1-69和Hpms2-281的鉴别能力较强。再用标记Hpms1-69和Hpms2-281的指纹数据分别与其他标记的指纹数据进行三标记组合,进行聚类分析,结果表明:约30%的三标记组合能鉴别参试杂交种,其中又以标记组合Hpms1-69+Hpms2-281+CA514272和Hpms1-69+Hpms2-281+BM61461为优。此外,15个SSR标记在12个辣椒杂交种中共检测到38个等位基因,出现两个杂交种指纹图谱完全相同的概率为3.63×10-12,表明这些标记可以用作目前构建辣椒指纹图谱的核心标记。

【Abstract】 Digital fingerprints of 12 capsicum hybrids in Guizhou Province were established by using 15 polymorphic SSR markers screened from 75 SSR markers through capsicum genome.Using the fingerprint data of each marker to do respective clustering analysis,the results showed the ability of identifying hybrids of the two SSR markers,Hpms1-69 and Hpms2-281.Furthermore,the fingerprints dada of three-marker combinations of Hpms1-69,Hpms2-281 and other markers were used to do clustering analysis,and the results showed that about 30% of three-marker combinations could identify the 12 hybrids,and two combinations of Hpms1-69+Hpms2-281+CA514272 and Hpms1-69+Hpms2-281+BM61461 had excellent identifying ability.In addition,38 alleles of the15 SSR markers were detected among 12 capsicum hybrids,and the probability that two hybrids had same fingerprint was 3.63×10-12.It indicates that the established fingerprints with the 15 markers can be used to identify other capsicum varieties besides the 12 hybrids,and the 15 SSR markers can be used as core markers for establishing other capsicum varieties fingerprint.
