牛Smad 9基因cDNA克隆及生物信息学分析
Cloning and Tissues Expression Analysis of cDNA Encoding the Bovine Smad 9 Gene
【摘要】 Smad 9属于R-Smads,在TGF-β信号传导过程中起着重要作用。本研究利用生物信息学方法,结合RT-PCR技术和SMARTRACE技术,得到全长为1 871 bp的牛Smad 9基因cDNA序列。通过核酸序列分析发现,牛Smad 9基因编码345个氨基酸残基,牛Smad 9蛋白无跨膜区和信号肽序列,该蛋白与人、小鼠、大鼠和红原鸡在氨基酸序列上分别有78.84%,79.21%,77.88%和70.38%的同源性。通过多组织RT-PCR分析发现,牛Smad 9基因的组织表达谱很广,在卵巢、肝、肌肉、小肠、脂肪、子宫、肾脏、心肌、肺、胰腺、睾丸、乳腺组织中均有表达。
【Abstract】 The Smad 9 is one of the members of R-Smads and played an important role in the signal transmission of TGF-β.The bovine Smad 9 gene cDNA was cloned by RT-PCR,RACE-PCR and got a 1871bp full-long cDNA sequence.The bovine Smad 9 gene codes 345 amino acids and shares 78.84%,79.21%,77.88% and 70.38%similarity in amino acid sequences with human,mouse,rat and jungle fowl respectively.The bovine Smad 9 had no signal peptide and transmembrane helix.Smad 9 mRNA was found in ovary,liver,skeleton muscle,small intestine,fat,uterus,kidney,heart,lung,pancreas,testis and galactophore by RT-PCR,which indicated Smad 9 was broad expressed in bovine.
