姜黄素对肺纤维化大鼠肺组织NF-κB和IκBα表达的影响
Effect of curcumin on the expression of NF-κB and IκBαin pulmonary fibrosis rats
【摘要】 目的观察姜黄素对博莱霉素诱导的肺纤维化大鼠肺组织NF-κB和IκBα表达的影响。方法 SD雄性大鼠144只,随机分为假手术组、模型组、醋酸泼尼松组及姜黄素高、中、低剂量组,每组24只。假手术组大鼠气管内一次性滴注等体积的生理盐水,余各组滴注盐酸博莱霉素。造模后第1天开始给药,持续到处死动物的前1天。各组动物于第7天、14天、28天随机处死8只,取肺组织分别采用免疫组化及Western-blot方法结合图像分析来检测各组大鼠肺组织中的NF-κB和IκBα表达水平。结果与假手术组相比,第7天、14天、28天时模型组大鼠肺组织中NF-κBp65的含量均显著升高(P<0.01),而IκBα的含量均显著降低(P<0.01);与模型组相比,醋酸泼尼松组及姜黄素高、中、低剂量均可降低大鼠肺组织中NF-κBp65的含量,而升高IκBα的含量,其中7 d、14 d时姜黄素中剂量组IκBα的含量升高较为显著(P<0.01),28d时,姜黄素高剂量组IκBα的含量升高最为显著(P<0.01)。结论一定剂量姜黄素能促进肺组织中IκBα的表达,而抑制NF-κB的活化和表达。
【Abstract】 objective To investigate the effect of curcumin on the expression of NF-κB and IκBα:in pulmonary fibrosis rats induced by bleomycin.Methods 144 male Sprague Dawley rats were randomly divided into 6 groups(24 rats in each group):Sham operation group,model group,prednisone acetate group,high curcumin dose group,moderate curcumin dose group,and low curcumin dose group.Bleomycin hydrochlorides were dropped into trachea in all groups except sham operation group,and same amount of saline were dropped into trachea in the sham operation group.Drugs were administered from the first day to the last day before the animals were killed.On 7d, 14d,28d,eight rats per group were randomly killed,the lung tissues were obtained to assess with immunohistochemical and Western blot method combined image analysis technique to detect NF-κB and IκBα:expression level.Results Compared with the sham group,the content of NF-κB P65 of the model group(7d,14d,28d) were significantly higher(P<0.01 ) whereas the content of IκBα:were significantly lower(P<0.01 );Compared with the model group,prednisone acetate group and curcumin(high,moderate,low dose) groups can reduce content of the NF-κB p65 in lung tissue,and increase the content of IκBα.Conclusion Curcumin can promote IκBαexpression and suppress NF-κB activation and expression in lung tissue.
