【摘要】 目的:观察垂盆草不同提取物的急性毒性反应。方法:经预实验,垂盆草水提物以最大浓度及最大给药体积的药液连续给小鼠灌胃2次,间隔7h,继续观察14d,记录小鼠毒副反应情况,计算最大耐受量及耐受量倍数;垂盆草醇提物按文献方法测定LD50。取小鼠50只随机分为5组,每组10只,以360g生药.kg-1(相当于临床单日剂量的600倍)为最高剂量,按1∶0.83的剂间比例,分别给5组小鼠灌胃给药2次,间隔7h(给药剂量分别为360,300,250,207,172g生药.kg-1)1次,常规饲养观察14d。记录小鼠毒性反应及死亡情况,按改进寇氏法公式进行计算。结果:垂盆草水提物的MTD为206.892g.kg-.1d-1,该剂量相当于人体推荐服用量344.82倍;垂盆草醇提物的LD50为208.305g.kg-1。结论:垂盆草提取物无明显毒性,认为临床常用量是安全可行的。更多还原

【Abstract】 Objective:To observe the acute toxicity reaction of different extracts from Sedum sarmentosum Bunge on mice.Methods:Through the pre-experiment,Mice were administered aqueous extract of Sedum sarmentosum Bunge by irrigating gastric(ig)with the highest concentration and the largest volume for 2 times,7 h interval,and were observed on the situation of acute toxicity in the successive 14 days,and to measure the maximum tolerance dose(MTD).Median Lethal Dose(LD50)of the ethanol extract of Sedum sarmentosum Bunge was measured by literature method.50 mice were randomly divided into five pharmaceutical dose groups,ten mice each group,set 360 g·kg-1(equivalent to clinical daily doses of 600 times)as the highest doses according to the 1∶ 0.83 between proportion,(Administration dosage respectively are 360,300,250,207,172 g·kg-1).Mice were administered alcohol extraction of Sedum sarmentosum Bunge by irrigating gastric(ig)for 2 times,7 h interval,and were observed on the situation of acute toxicity in the successive 14 days.Toxicity and death records of mice are measured by improved Karber method.Results:The maximum tolerance(MTD)dose of aqueous extract of Sedum sarmentosum Bunge after the acute toxic experiment was not less than 206.892 g·kg-1·d-1,which was equal to 344.82 times of aducts’ oral dose per day.the LD50 of the alcohol extraction of Sedum sarmentosum Bunge is 208.305 g·kg-1.Conclusion:Extracts from Sedum sarmentosum Bunge have little acute toxic in mice.They are relatively safe.更多还原
