芩丹胶囊对高血压大鼠血管外膜重构的影响

【摘要】 目的探讨芩丹胶囊对自发性高血压大鼠(SHR)血管外膜重构的影响。方法32只40周龄老年自发性高血压大鼠(SHR)随机分为模型(C)组、芩丹胶囊大剂量(D)组、芩丹胶囊小剂量(E)组、氯沙坦(F)组,另设老年Wistar大鼠空白对照(A)组及芩丹胶囊正常用药(B)组。观测各组大鼠收缩压;灌胃结束后取胸主动脉HE染色观察外膜细胞核密度变化;激光共聚焦显微镜观测胸主动脉内径,中、外膜比例;免疫组化法及实时定量PCR检测转化生长因子(TGF-β1)在胸主动脉外膜的表达。结果与C组相比,D、E组收缩压均有了明显下降;胸主动脉外膜细胞核密度减少;胸主动脉内径/中膜/外膜的比例明显升高,TGF-β1在外膜表达降低。结论芩丹胶囊不仅能够有效地降低SHR血压,而且能够对动脉血管外膜重构具有明显地抑制及逆转作用。其机制可能与下调血管外膜TGF-β1合成、分泌,抑制成纤维细胞(AF)向具有平滑肌细胞特征的肌成纤维细胞(MF)转换,进而抑制血管外膜重塑有关。

【Abstract】 Objective To investigate the effects of qindan capsule(QC)on vascular adventitial fibroblasts remodling in spontaneously hypertensive rats(SHRs).Methods Thirty-two SHRs of 40 weeks old were divided into the model(group C),high dose QC(group D),low dose QC(group E),losartan(group F).Sixteen Wistar rats were taken as the normal controls(group A)and QC group(group B).Blood pressure was recorded every three weeks.After 12 weeks,the thoracic aorta in groups of D,E and F was taken for observing the morphological changes and nuclear density by HE staining and laser scanning confocal microscope,and detecting the mRNA expression of transforming growth factor β1(TGF-β1)in thoracic aorta by immunohistochemical staining and real-time quantitative fluorescent PCR.Results Compared with group A,QC could reduce the blood pressure of SHRs,increase the ratio of ID/mesolamella/adventitia of endangium and the expression of TGF-β1.Conclusion QC could not only reduce blood pressure of SHRs,but also suppress and even reverse adventitial vascular remodeling.The mechanism may be related to the down-regulation of TGF-β1 mRNA expression in thoracic aortic wall and inhibition of myofibroblasts transformation.
