胰岛素样生长因子结合蛋白-4在围植入期小鼠子宫内膜的表达及意义

【摘要】 目的检测胰岛素样生长因子结合蛋白-4(IGFBP-4)在围植入期小鼠子宫内膜的表达分布及其生物学作用。方法取未孕动情期、真孕及假孕3~6d小鼠子宫做切片,采用免疫组化SP法检测IGFBP-4在小鼠子宫内膜的表达情况。结果 IGFBP-4在小鼠子宫内膜主要分布于腺上皮细胞、腔上皮细胞、基质细胞及蜕膜细胞。IGFBP-4在真孕组小鼠子宫内膜于孕3d出现阳性表达;孕4~5d中等强度表达;孕6d表达最强,主要分布于整个蜕膜区细胞。未孕动情期小鼠子宫内膜IGFBP-4呈弱表达。假孕组小鼠子宫内膜IGFBP-4只在孕5~6d有微弱表达。真孕组小鼠子宫内膜IGFBP-4表达明显强于同期假孕组。结论 IGFBP-4在围植入期小鼠子宫内膜持续表达,提示IGFBP-4可能在小鼠胚泡植入过程发挥重要生物学作用。

【Abstract】 Objective To observe the expression and distribution of insulin-like growth factor binding protein-4 (IGFBP-4) and its biological effects in the peri-implantation mouse endometrium.Methods Uterus of mice in the nonpregnant (estrus stage) ,pregnancy and pseudopregnant 3 to 6 days were obtained.Immunohistochemical SP staining method was used for detecting the expression of IGFBP-4 in the endometrium during mice embryo impantation.Results The expression of IGFBP-4 was distributed in the glandular epithelium,luminal epithelium,stromal cells and decidual cells.In the pregnant group,the expression of IGFBP-4 in the uterine endometrium on the 3rd day began to appear,increased in 4th day and 5th day,peaked at the 6th day,mainly distributed throughtout decidua cells.In the pseudopregnant group,weak expression of IGFBP-4 was only observed on the 5 th to 6th days of pregnancy.The expression of IGFBP-4 was weakly in the endometrium of nonpregnant mice.The expression of IGFBP-4 mice in endometrium of the pregnant group was significantly higher than that in the homeochronous pseudopregnancy group.Conclusion IGFBP-4 expresses continuously in mouse endometrium during day 3 ~ 6 of pregnancy,It suggests that IGFBP-4 might play an important biological role in the process of the mouse embryo implantation.
