【摘要】 目的在以往研究的基础上,改进早孕期绒毛短期培养后染色体制备的方法。方法采用的培养液血清浓度为5%,绒毛细胞经过48 h培养,将酶解离细胞、低渗处理与高浓度秋水仙素处理简化为一步,使用冰醋酸分步解离。结果染色体分散好,带型清晰。结论该技术简单省时,易于操作,成功率较高,便于临床推广使用。更多还原

【Abstract】 Objective A modified technique of chromosome preparation from short-term culture of villi was established upon previous studies.Methods The chorionic villi were cultured for 48 hours at a medium serum concentration of 5%,to merge cell enzymolysis separation from the cytotrophoblastic layer,treatment with hypotenic solution and high concentrations of colchicines treatment into one step,i.e.,step-by-step dissociation by glacial acetic acid.Results The chromosomes were evenly distributed with clear banding pattern.Conclusion The technique is simple,time-saving and easy to operate,with a high success rate,which facilitates clinical popularized application.更多还原
