前胡丙素对缺氧再灌注损伤心肌细胞的保护作用
Protective effects of praeruptorin C On cardiomyocytes induced by hypoxia-rexoygenation induced injury
【摘要】 目的研究前胡丙素对培养心肌细胞缺氧再灌注损伤的保护作用。方法采用SD大鼠乳鼠进行心肌细胞培养,建立模拟缺氧再灌注模型。实验分4组:正常组(contro1):细胞正常培养;缺氧预适应组(HP):预先缺氧2 h,复氧1 h,再缺氧12 h后,复氧2 h;前胡丙素预处理组(PP):先予前胡丙素单体终浓度为100 mol/L作用1 h,再行缺氧12 h,后复氧2 h;模拟缺氧再灌注组(HR):缺氧12 h,后复氧2 h。观察心肌细胞搏动频率及细胞形态;细胞上清检测乳酸脱氢酶(LDH)、肌酸磷酸激酶(CK)值。结果①细胞博动率:前胡丙素能增加缺氧再灌注损伤心肌细胞搏动频率;②细胞形态:心肌细胞H/R培养后皱缩、变圆,伪足减少,PP组心肌细胞形态变化小于对照组;③LDH和CK值:与正常培养组比较,模拟缺氧再灌注组细胞上清中LDH、CK值明显升高(P<0.01);与模拟缺氧再灌注组比较,HP组、PP组细胞上清中LDH、CK值明显降低(P<0.01);但前胡丙素预处理组与缺氧预适应组之间无明显差异(P>0.05)。结论前胡丙素能保护缺氧再灌注损伤心肌细胞,减轻损伤程度。
【Abstract】 Objective To investigate the protective effect of praeruptorin C on cardiomyocytes induced by hypoxia/rexoygenation(H/R)induced injury.Methods The primary cadiomyocytes were randomly divided into 4 groups: the normally cultured group(contro1),the hypoxia-preconditioning(HP) group,the praeruptorin C preconditioning(PP) group and the hypoxia/rexoygenation(HR)group.Each group was untreated or treated with 100 mol/L praeruptorin C.At the end of the experiment,cadiomyocytes beating rate and morphology were observed;The value of LDH and CK of celluar supematant were detected by automatic biochemistry analyzer.Result ① the beating rate of cardiomyocytes: praeruptorin C increased beating rate of cardiomyocytes induced by hypoxia/rexoygenation(H/R);② morphology: after H/R injury,cardiomyocytes contracted,tending to get ground in shape and its pseudopods decreased,while marked morphological changes were not observed in PP group;③ the value of LDH and CK: compared with the contro1 group,LDH and CK content increased significantly in HR group(P<0.01);compared with HR group,LDH and CK contents receded in HP and PP group(P<0.01),but LDH and CK contents of PP group was similar to that of HP group(P>0.05).Conclusion praeruptorin C might be beneficial for the protection of cardiomyocytes followed by hypoxia/rexoygenation because of the attenuation of oxidative damage.
