七氟甲醚菊酯亚急性吸入毒性实验研究

【摘要】 目的测定七氟甲醚菊酯大鼠28 d吸入毒性。方法选择无特定病原体级成年SD大鼠,随机分成4组,每组12只,雌雄各半,分别吸入七氟甲醚菊酯0、0.58、1.70和6.48 mg/m3,每天4 h,每周5 d,共4周,实验结束时取血液分析丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)等11项生化指标,白细胞计数(WBC)、红细胞计数(RBC)等9项血常规指标,取心、肝、脾等7种主要脏器称重并做病理学检查。结果七氟甲醚菊酯1.70和6.48 mg/m3组动物染毒期间出现轻微震颤、口鼻周有血性分泌物等中毒表现。与对照组比较,1.70和6.48 mg/m3组雌性动物血液中WBC减少(P<0.01);6.48 mg/m3组动物体质量下降(P<0.05),其中雄性动物嗜碱粒细胞百分率降低(P<0.05)、肾脏系数增大(P<0.05),雌性动物血清碱性磷酸酶(AKP)水平高(P<0.05)。结论七氟甲醚菊酯对SD大鼠的28 d亚急性吸入最大无作用质量浓度为0.58 mg/m3(4 h/d)。

【Abstract】 Objective To determine the subacute toxicity of [2,3,5,6-Tetrafluoro-4-(methoxymethyl)phenyl] methyl 2,2-dimethyl-3-(3,3,3-trifluoro-1-propenyl) cyclopropanecarboxylate(TPMDC) inhaled by rats for 28 days.Methods SPF SD rats were randomly divided into 4 groups with 12 rats in each group,6 females and 6 males.The animals inhaled with TPMDC at the concentration of 0,0.58,1.70 and 6.48 mg/m3 for 4 weeks(4 h/d,5 d/w),respectively.At the end of the test,blood samples were collected for analysis of complete blood count and serum biochemical indexes.The main organs were weighed and pathological changes were observed.Results In both medium and high concentration groups(1.70 and 6.48 mg/m3) exposed to TPMDC,poisoning symptoms were observed including trembling,blood-like-secretion around mouth and nose.Compared with the control group,it was found that white blood cell(WBC) of female rats decreased.Body weight decreased in high concentration group,while the percentage of basophilic granulocyte reduced,kidney ratio(kidney weight/body) increased in male rats and AKP level increased in female rats.Conclusion The results suggested that the non-observed adverse effect level(NOAEL) of TPMDC in SD rats was 0.58 mg/m3(4 h/d for 28 days).
