玉郎伞2种查尔酮单体对心肌细胞凋亡的抑制作用

【摘要】 目的研究从玉郎伞中首次分离的2种查尔酮单体2,6-二甲氧基查尔酮(YLSA)和17-甲氧基-7-羟基-苯骈呋喃查尔酮(YLSB)对心肌细胞凋亡的作用。方法原代培养大鼠心肌细胞,利用终浓度为100μmol·L-1的过氧化氢孵育16h建立凋亡模型;ELISA法测定心肌细胞培养上清液中TNF-α含量,Annex V-FITC/PI双染法检测心肌细胞凋亡,免疫组化法检测NF-κBp65、Bcl-2、Bax蛋白的表达。结果YLSA和YLSB均可抑制心肌细胞凋亡,降低TNF-α活性,下调NF-κBp65和Bax蛋白表达,上调Bcl-2蛋白表达,增加Bcl-2/Bax比值。结论YLSA和YLSB具有抑制心肌细胞凋亡的作用。

【Abstract】 AIM To observe the effects of two chalcone compounds firstly separated from Yulangsan on cardiocyte apoptosis. METHODS The apoptosis model of cultured neonatal rat cardiomyocytes was developed by adding 100 μmol·L-1 H2O2 as final concentration into cell culture fluids for 16 h. The contents of TNF-α was determined by ABC-ELISA, while the apoptosis rate of the cardiocytes was detected by Annex V-FITC / PI double staining. The protein expressions of NF-κBp65, Bcl-2 and Bax were evaluated by using immunohisto-chemical method. RESULTS Compared with the model group, preconditioning by two chalcone compounds could decrease the apoptosis rate and the contents of TNF-α (P < 0.05) . Meanwhile, two chalcone compounds down-regulated the protein expressions of NF-κBp65 and Bax, as well as up-regulated the protein expressions of Bcl-2 and the ratio of Bcl-2 / Bax (P < 0.05 vs. model) . CONCLUSION Two chalcone compounds have potent effects on relieving cardiocyte apoptosis.
