【摘要】 目的:探讨化纤灵方药物血清对肝星状细胞(HSC)增殖及凋亡的影响。方法:传代培养的大鼠HSC系HSC-T6与中药复方化纤灵方药物血清(5%、10%、20%)共同培养48h后,应用MTT法检测HSC-T6细胞增殖,琼脂糖凝胶电泳法及流式细胞仪测定细胞凋亡。结果:复方化纤灵方药物血清能显著抑制体外培养的HSC-T6增殖,中、高剂量组凋亡率分别为(20.37±0.78)%、(21.69±2.35)%、显著高于正常对照组(P<0.01);化纤灵方药物血清均可诱导HSC凋亡。结论:复方化纤灵方能抑制HSC增殖并促进其凋亡,可能是抗肝纤维化作用的主要机制。更多还原

【Abstract】 Objective:To investigate the effect of Chinese herb Compound Huaxianling on apoptosis and proliferation of HSC-T6.Methods:HSC-T6 was introduced into our laboratory and subcultured.Normal rats were medicated with Compound Huaxianling and the serum was collected to prepare the medicated serum(5%,10%,20%),which was incubated with the subcultured HSC-T6.Then,to observe the proliferation of HSC-T6 by MTT colorimetric assay and detect the cell apoptosis rate by using flow cytometer and agarose gel electrophoresis.Results:Compound Huaxianling could significantly inhibit HSC-T6 proliferation in vitro,and the apoptosis rate(%) of middle-dose and high-dose were 20.37±0.78 and 21.69±2.35,respectively,which were significantly higher than that of the blank-control group(P<0.01).Conclusion:Compound Huaxianling could significantly inhibit the proliferation and accelerate the apoptosis of HSC,which may be the main mechanism of Huaxianling on anti-hepatic fibrosis actions.更多还原
