EGCG对肾小球系膜细胞MMP-9和细胞外基质合成的影响

【摘要】 目的探讨表没食子儿茶素没食子酸酯(EGCG)对脂多糖(LPS)诱导的肾小球系膜细胞(GMCs)基质金属蛋白酶系统和细胞外基质合成的影响。方法以大鼠GMCs为实验对象,分为正常对照组、LPS组、LPS+EGCG组。CCK细胞计数法检测EGCG作用下大鼠GMCs的OD值,酶联免疫吸附实验(ELISA)检测基质金属蛋白酶-9(MMP-9)及抑制物金属蛋白酶组织抑制因子-1(TIMP-1)蛋白表达,并检测细胞外基质成分纤维连接蛋白(FN)、Ⅳ型胶原(ColⅣ)及层粘连蛋白(LN)的含量。结果LPS诱导TIMP-1蛋白和细胞外基质合成增加,与正常对照组比较差异有统计学意义(P<0.05或P<0.01),EGCG能抑制升高TIMP-1蛋白表达和细胞外基质合成。而各组MMP-9蛋白表达变化不明显。结论EGCG可抑制LPS诱导的大鼠GMCs增殖和细胞外基质的合成,为EGCG的肾脏保护作用提供理论依据。

【Abstract】 Objective To explore the effects of epigallocatechin-3-gallate on expression of matrix metalloproteinase-9(MMP-9) and extracellar matrix production(EMC) induced by lipopolysaccharide(LPS) in glomerular mesangial cells(GMCs).Methods GMCs were divided into normal control group,LPS group and EGCG+LPS group.The growth inhibiting effect of EGCG on mesangial cells was measured by CCK-8 method.The levels of MMP-9 and EMC in the supernatants were determined by enzyme-linked immunosorbant assay(ELISA).Results CCK-8 methods showed that LPS stimulated proliferation of GMCs,and this proliferation could be inhibited by EGCG in a time-dependent manner.The content of MMP-9 in every group was not different.The content of tissue inhibitor of metalloproteinase-1(TIMP-1) in LPS group was obviously higher than that in control group.EGCG could ameliorate the levels of FN,ColⅣand LN of the rat mesangial cells induced by LPS.Conclusion EGCG can significantly inhibit proliferation and secretion of TMP-1 and EMC in LPS-induced rat mesangial cells,and provides the theoretical evidence for the renal protection of EGCG.
