【摘要】 目的探讨大鼠β₃肾上腺素能受体（β₃受体）表达与左心室肥厚的关系。方法 40只雄性Wistar大鼠随机分为假手术组（20只）和研究组（20只）,研究组采用主动脉缩窄法建立压力负荷性心肌肥厚大鼠模型。两组分别于造模后4周、8周检测超声心动图指标、左心室质量指数及RT-PCR方法测定左心室β₃受体的mRNA表达。结果与假手术组比较,研究组大鼠造模后第4周左心室后壁舒张末期厚度（INPWTD）及室间隔舒张末期厚度（TVSTD）明显增加（P<0.05）,但左心室舒张末期内径（LVEDD）无明显变化（P>0.05）。研究组大鼠造模后第8周与第4周比较,LVPWTD及IVSTD增加得更明显（P<0.01）,LVEDD显著缩小（P<0.01）。造模后第4周、第8周研究组大鼠左心室质量指数均较假手术组显著增加（P<0.01）,且造模后第8周增加得更显著（P<0.01）。假手术组造模后第4周、第8周大鼠左心室β₃受体mRNA表达之间比较差异无统计学意义[（0.595±0.052）比（0.602±0.050）,P>0.05]。研究组大鼠左心室β₃受体表达较假手术组明显升高（P<0.01）,造模后第8周较第4周升高的更显著[（1.240±0.036）比（1.182±0.029）,P<0.01]。无论假手术组还是研究组,左心室β₃受体mRNA表达与INMI均呈明显的正相关。结论大鼠肥厚左心室的β₃受体表达明显增加,且表达水平与左心室肥厚的程度相关。更多还原

【Abstract】 Objective To investigate the relationship between β₃-adrenoceptor expression and left ventricular hypertrophy （LVH） in rats.Methods Forty male Wistar rats were randomly divided into sham-operated group（sham group） and study group,twenty rats in each group.The left ventricular hypertrophy model was established by abdominal aortic constriction.Echocardiographic parameters,left ventricular mass index （LVMI） and the level of β₃-adrenoceptor expression were analyzed in the two groups four weeks and eight weeks after operation.Results The left ventricular posterior wall thickness at end diastole （LVPWTD） and interventricular septum thickness at end diastole（IVSTD） in the study group were thicker than in the sham group （P<0.05）,but the left ventrieular end-diastolic dimension （LVEDD） was similar between the two groups （P>0.05） four weeks after operation.In the study group,LVPWTD and IVSTD were significantly increased （P<0.01）,and LVEDD was significantly decreased （P<0.01） eight weeks after operation compared with those four weeks after operation （P<0.01）.The LVMI in the study group was higher than in the sham group（P<0.01） after operation.In the study group,the LVMI was significantly increased （P<0.01） eight weeks after operation compared with those four weeks after operation.The level of β₃-adrenoceptor mRNA expression in left ventricle in the study group was significantly higher eight weeks after operation compared with those four weeks after operation （1.240 ±0.036 vs.1.182±0.029,P<0.01）,whereas it remained unchanged in the sham group.In each group,there was a significant correlation between the level of β₃-adrenoceptor expression and the LVMI.Conclusion There is a significant increase in the β₃-adrenoceptor expression in the hypertrophic ventricles in rats.The increase in β₃-adrenoceptor expression is related to the severity of ventricular hypertrophy.更多还原
