中波紫外线诱导小鼠表皮细胞Bax和Bcl-2的表达

【摘要】 [目的]探讨中波紫外线(UVB)对正常小鼠表皮细胞凋亡及凋亡相关基因Bax和Bcl-2的调控。[方法]将正常ICR小鼠随机分为对照组(假照射组)、500J/m2、1000J/m2、1500J/m2、2000J/m2紫外线照射后2h组(每组10只小鼠)。1500J/m2紫外线照射后4h组、8h组、12h组、24h组、48h组、72h组(每组10只小鼠)。采用链霉菌亲生物素蛋白-过氧化物酶标免疫组织化学法(streptavidin-peroxidase,SP)染色检测凋亡相关基因Bax和Bcl-2的蛋白表达。[结果]小鼠皮肤用不同剂量紫外线照射2h后Bax的表达,随紫外线剂量增大而变大,Bcl-2表达减小,呈剂量依赖性。用1500J/m2紫外线照射小鼠后12hBax表达达到高峰,Bcl-2表达减弱;24h后Bax表达开始减弱,而Bcl-2则开始表达,72h恢复正常,Bax和Bcl-2的表达与紫外线照射后时间有关。[结论]中波紫外线照射小鼠皮肤后,Bax表达上调,Bcl-2表达下调,促进小鼠紫外线损伤的表皮细胞凋亡。

【Abstract】 [Objective] To investigate regulative effects of ultraviolet B(UVB) for normal apoptosis of epidermal cell in mice and apoptosis-related gene Bax and Bcl-2.[Methods] Normal ICR mice were divided into control group(sham irradiation group) and ultraviolet irradiation groups with dosage of 1 000 J /m2,1 500 J /m2,2 000 J /m2 for 2 hours,and 1 500 J /m2 for 4,8,12,24,48,72 hours groups with 10 mice each group.Protein expression of apoptosis-related gene Bax and Bcl-2 was detected by immunohistochemical method with streptavidin-peroxidase.[Results] Bax expression increased and Bcl-2 expression decreased in mouse skin after UVB irradiation with different dosages for 2 hours and both appeared dosage-dependant.Bax expression increased to peak value after UVB irradiation with 500 J /m2 dosage for 12 hours whereas Bcl-2 expression decreased.Bax expression began to decreased and Bcl-2 expression increased after 24 hours,and returned to normal value after 72 hours.Bax expression and Bcl-2 expression were related to time after UVB irradiation.[Conclusion] Bax expression increases and Bcl-2 expression decreases in mouse skin after UVB irradiation,so that it promotes apoptosis of epidermal cells injured by ultraviolet.
