乌斯他丁对乳腺癌细胞MCF-7、MDA-MB-231MMP-9的表达及增殖侵袭的影响

【摘要】 [目的]探讨乌斯他丁对体外培养的乳腺癌细胞MCF-7(雌激素受体阳性)、MDA-MB-231(雌激素受体阴性)MMP-9(基质金属蛋白酶)的表达及增殖侵袭的影响。[方法]将体外培养的乳腺癌细胞MCF-7和MDA-MB-231分别分为4组:对照组、UTI高、中、低剂量组,分别用相应药物处理。分别检测两种细胞的增殖能力、浸袭能力、MMP-9基因的表达。[结果]乌斯他丁明显抑制MCF-7和MDA-MB-231细胞的增殖侵袭能力,并能使两种细胞中MMP-9基因的转录水平下降。[结论]乌斯他丁能抑制乳腺癌细胞的增殖、侵袭作用,并能降低两种细胞MMP-9基因的表达。
【Abstract】 [Objective] To investigate effect of Ulinastatin on cultured breast cancer cells MCF-7 (estrogen receptor positive), MDA-MB-231 (estrogen receptor negative) MMP-9 (matrix metalloproteinase) expression and the proliferation and invasion. [Methods] Cultured MCF-7 breast cancer cells and MDA-MB-231 were divided into four groups: control group, UTI high, medium and low dose group, all were treated with appropriate medication. Detected the proliferation of two cells, invasion capabilities, MMP-9 gene expression. [Results] Ulinastatin inhibited MCF-7 and MDA-MB-231 cell proliferation and invasion, and made MMP-9 gene transcription levels in two kinds of cells decrease. [Conclusion] Ulinastatin can inhibit breast cancer cell proliferation, invasion, and reduce the MMP-9 gene expression in two kinds of cells.
