原发性高血压患者ACE水平与基因多态性

【摘要】 [目的]探索浙江省畲汉两族原发性高血压患者血清血管紧张素转换酶(ACE)水平与ACE基因多态性的关系。[方法]采用聚合酶链反应(PCR)检测215例畲汉两族原发性高血压(病例组)及正常人群血中ACE基因16内含子I/D多态标记;采用紫外分光光度法测定血清ACE水平;运用多元线性回归分析血清ACE水平与ACE基因型的关系。[结果]浙江畲汉两族血清ACE水平均为病例组高于对照组(P﹤0.001)。在两族的病例组和对照组中,不同ACE基因型之间血清ACE水平的差别有统计学意义(P﹤0.001),两两比较,各组均为DD型﹥ID型﹥II型。多元线性回归显示DD基因型和ID基因型相对于II基因型能使血清ACE水平增加(P﹤0.001)。[结论]原发性高血压患者血清ACE水平较对照组高;畲汉两族的民族因素与血清ACE水平无关;两族中DD及ID基因型的个体血清ACE水平较II基因型高。

【Abstract】 [Objective] To explore the essential hypertension (EH) patients’ serum angiotensin-converting enzyme (ACE) level and its association with the gene polymorphism of ACE in She and Han nationalities of Zhejiang province. [Meth- ods] The I/D polymorphic markers of intron 16 of ACE gene were detected in 215 EH and normal individuals of She and Han nationalities by using polymerase chain reaction (PCR), ultraviolet spectrophotometric method was used to detect the serum ACE level, multiple linear regression was used to analyze the association between serum ACE level and ACE genotypes. [Results] The serum ACE levels in EH groups were higher than that in control groups in both She and Han nationalities (P﹤ 0.001). In EH and control groups, there were statistical significances between different genotypes and serum ACE (P﹤0.001), the order was DD﹥ID﹥II in all groups. Multiple linear regression indicated that DD and ID genotypes can increase the serum ACE level compared with II genotype. [Conclusion] EH petients’ serum ACE level is higher than that in control group. The serum ACE level is not associated with the She and Han nationalities. Individuals with DD and ID genotypes have higher ACE level in She and Han nationalities.
