原发性肝癌患者外周血中IL-8、IP-10及其mRNA表达
EXPRESSION OF IL-8,IP-10 AND ITS MRNA IN SERUM AND PBMC OF THE PATIENTS WITH PRIMARY HEPATIC CARCINOMA
【摘要】 [目的]探讨原发性肝癌患者外周血IL-8、IP-10及其mRNA表达,局部肿瘤手术切除对其表达的影响。[方法]选择2006年1月~2006年12月安徽理工大学附属医院、淮南新华医院收治的原发性肝癌患者64例,选取健康人群70例为正常对照组。以酶联免疫吸附检测患者血清中IP-10、IL-8蛋白含量;荧光实时定量PCR检测患者外周血单个核细胞内IP-10mRNA、IL-8mRNA含量。[结果]原发性肝癌患者血清IL-8、IP-10水平分别为(486.2±52.3)pg/ml、(386.7±42.5)pg/ml;PBMC内IL-8mRNA、IP-10mRNA水平分别为(1.1352±0.1395)、(0.6387±0.0768),显著高于正常对照组(P﹤0.01);22例原发性肝癌患者第1周血清IL-8、IP-10及其mRNA水平应激性升高,术后1周血清IL-8、IP-10分别为(869.6±90.7)pg/ml、(439.8±51.5)pg/ml;PBMC内IL-8mRNA、IP-10mRNA水平分别为(1.3671±0.1447)、(0.8032±0.0817),与术后第1个月、第3个月比较差异有统计学意义(P﹤0.01)。[结论]原发性肝癌患者外周血中IL-8、IP-10及其mRNA表达高于正常对照组,局部肿瘤切除手术后1周内导致外周血IL-8、IP-10及其mRNA认识应激性升高,其随术后恢复而逐渐下降。
【Abstract】 [Objective] To study the expression of interleukin-8 (IL-8), interferon γ-inducible protein l0 (IP-10) and its mRNA in serum and peripheral blood mononuclear cells (PBMC) of the patients with primary hepatic carcinoma (PHC), and investigate the effect of tumorectomy on the expression. [Methods] A total of 64 cases with primary hepatic carcinoma (PHC) from affiliated hospital of Anhui University of Science and Technology and Huainan Xinhua Hospital form January 2006 to December 2006 were chosen, and 70 healthy people were chosen as control group. Typical PHC patients were selected. The level of IP-10 and IL-8 were detected by ELISA serum, respectively, IL-8 mRNA and IP-10 mRNA were measured by Real-time PCR in PBMC. The level of complementary DNA against GAPDH (lgcDNA/lgGAPDH) was regarded as the extreme level of chemokine. [Results] The levels of IL-8 and IP-10 in serum of PHC patients were (486.2 ± 52.3) pg/ml and (386.7 ± 42.5) pg/ml, respectively. The levels of IL-8 mRNA and IP-10 mRNA in PBMC of PHC patients were 1.135 2 ± 0.139 5 and 0.638 7 ± 0.076 8, respectively. The levels of IL-8 and IP-10 and its mRNA were significantly higher than those in control group (P﹤0.01). The levels of IL-8 and IP-10 and its mRNA increased in 22 cases of patients with PHC after operation because of irritability. The levels of IL-8 and IP-10 in serum and its mRNA in PBMC one week after operation were (869.6 ± 90.7) pg/ml and (439.8 ± 51.5) pg/ml, (1.367 1 ± 0.144 7) and (0.803 2 ± 0.081 7), respectively, which were significantly higher than those of other Postoperative groups (P﹤0.01). [Conclusion] The expression of IP-10, IL-8 and their mRNA in serum of the cases of patients with PHC is significantly higher than those of control group, local tumor excision leads to the rise of expression because of irritability, which decreases gradually after tumor excision
