大豆皂甙对矽肺纤维化大鼠肺组织细胞Fas/FasL系统表达的影响

【摘要】 [目的]探讨大豆的皂甙(Soyasaponin,SS)对实验性大鼠矽肺纤维化模型中肺组织细胞Fas/FasL表达的影响。[方法]通过复制大鼠矽肺纤维化模型,观察各组大鼠血清中超氧化物歧化酶(SOD)、肺组织丙二醛(MDA)在染尘及SS干预后的含量变化。采用免疫组化法观察各组大鼠肺组织细胞Fas和FasL表达的改变。[结果]给予SS后,与对模型组相比血清SOD升高、MDA显著降低(P﹤0.05)。SiO2组Fas蛋白表达为相应对照组的4.51和5.05倍,其阳性信号主要集中于支气管上皮细胞、肺泡上皮细胞。[结论]Fas/FasL系统可能参与矽肺纤维化的发病过程,一定剂量的大豆皂甙可能通过降低氧化应激,从而缓解矽肺纤维化的进展。

【Abstract】 [Objective] To discuss the effect of SS on expression of Fas and FasL of Pneumonocyte in the Silicotic Fibro-sis Rats. [Methods] Superoxide dismutase (SOD) and pulmonary tissue malondialdehyde (MDA) in lung and their changes after SS intervened were observed. The immunohistochemical method was used to detect the level in the expression of Fas / FasL protein. [Results] Compared with NS group,the level of SOD decreased constantly in SiO2 group (P﹤0.05). MDA was increased significantly and maintained the high level (P﹤0.05). Expression of Fas protein of SiO2 group was 4.51 and 5.05 times compared with its NS group. Its positive staining was located in epithelial cells of bronchi-duct,alveolar and vascular endothelial cells. [Conclusion] Fas and FasL system might take part in the pulmonary fibrosis. A certain dose of SS might down regu-late the expressions of Fas and FasL to relieve the development of pulmonary fibrosis.
