MCP-1对成骨前体细胞凋亡及凋亡调控蛋白Caspase3表达的影响
The influence of MCP-1 on the apoptosis of osteoblast precursors and the expression of apoptosis Caspase3 protein
【摘要】 目的探讨MCP-1对成骨细胞凋亡和凋亡调控蛋白Caspase3表达的影响。方法体外培养小鼠MC3T3-E1细胞并采用免疫荧光方法检测其趋化因子MCP-1的表达情况,MC3T3-E1细胞加入不同浓度趋化因子MCP-1,流式细胞技术检测细胞凋亡率同时免疫细胞化学染色测定凋亡调控蛋白Caspase3的表达。结果 MC3T3-E1细胞MCP-1阳性表达。流式细胞仪检测结果表明,正常血清对照组存在小鼠MC3T3-E1细胞自发性凋亡,但均不大于5%。与正常血清组相比,无血清对照组细胞凋亡率明显升高。与无血清对照组相比,(1~100)ng/mL MCP-1组的浓度范围内,细胞凋亡率明显下降(P<0.05)。MCP-1组Caspase3平均灰度值降低(P<0.05),且随着MCP-1(1ng/mL~100ng/ml)浓度的增大,Caspase3平均灰度值呈下降趋势。结论 MCP-1可能通过调节Caspase3在成骨细胞中的表达,对抗无血清诱导的细胞凋亡,从而促进成骨的形成。
【Abstract】 Objective To investigate the influence of MCP-1 on the apoptosis of osteoblast precursors and to evaluate the expression of apoptosis Caspase3 protein.Methods The expression of chemotactic factor MCP-1 in MC3T3-E1 was detected by immunofluorescence in vitro.Different concentrations of chemotactic factor MCP-1 were added in MC3T3-E1 cells,the apoptosis rate was detected by flow cytomertry and the expression of Caspase3 was checked by immunocytochemistry.Results The expression of MCP-1 was positive in MC3T3-E1.Flow cytomertry test declared that the apoptosis of MC3T3-E1 with serum culture medium was not more than 5%.Compared with normal serum group,the apoptosis rate of serum-free group increased significantly.In the range of(1~100)ng/mL MCP-1,the MCP-1 apoptosis was declined compared with the serum-free group(P<0.05).Caspase3 was reduced on average gray values in MCP-1 group(P<0.05).With the concentration of MCP-1(1ng/mL~100ng/ml) increased,the expression of Caspase3 was declined.Conclusion That MCP-1 regulated the expression of Caspase3 in MC3T3-E1 resisted possibly the apoptosis induced by serum-free,thus promoted osteogenesis.
