Midkine在胰腺癌诊断中的作用研究
Study on Expression of Midkine in Pancreatic Cancer Diagnosis
【摘要】 目的:通过对胰腺肿瘤组织和对应的癌旁正常组织的Midkine(MK)mRNA定量检测、比较分析,为胰腺癌的早期诊断建立分子标志。方法:收集胰腺癌组织及癌旁组织人体标本,冷冻保存;提取组织总RNA。根据MK两端序列设计特异性引物,同时用β-肌动蛋白作为对照基因,逆转录PCR(RT-PCR)和荧光定量逆转录PCR(qRT-PCR)对MKmRNA分别进行定性和定量测定。结合统计学分析结果,判断分析MKmRNA含量和胰腺癌的相关性。结果:胰腺癌组织中MKmRNA表达明显高于癌旁组织(p<0.05)。结论:基于与胰腺癌的相关性,MK有可能作为胰腺癌早期诊断的分子标记。
【Abstract】 Objective:To establish a molecular marker for the earlier diagnosis of pancreatic cancer,We investigated the feasibility of quantitative analysis of Midkine(MK)mRNA in pancreatic cancer tissues and non-cancerous tissue.Methods:Total RNA was isolated from human pancreatic cancer and normal paracancerous pancreas tissue,MK mRNA was detected by RT-PCR and real-time PCR,β-actin was used as a reference gene.Results:The levels of expression of Midkine in pancreatic cancer tissue were significant higher than that in normal tissue.Conclusion:Based on the relativity with pancreatic cancer,Midkine could be a possible molecular marker for the earlier diagnosis of pancreatic cancer.
