金属离子对褐黄孢链霉菌生长和纳他霉素生物合成的影响
Effects of metal ions on growth of Streptomyces gilvosporeus and biosynthesis of natamycin
【摘要】 【目的】研究不同金属离子对褐黄孢链霉菌菌体生长和纳他霉素生物合成的影响。【方法】用单因素摇瓶发酵试验方法,研究不同浓度Na+,Fe2+,Mg2+,K+,Ca2+,Cu2+,Mn2+,Zn2+和Co2+对褐黄孢链霉菌生长和纳他霉素生物合成的影响。【结果】在发酵培养基中添加Mg2+和Ca2+可促进褐黄孢链霉菌的菌体生长,添加Fe2+、Cu2+、Mn2+和Co2+可抑制菌体生长,而添加Na+、K+和Zn2+对菌体生长无显著影响;在发酵培养基中添加低浓度K+、Ca2+和Co2+可促进纳他霉素的生物合成,添加Na+、Fe2+、Cu2+和Zn2+抑制纳他霉素的生物合成,而添加Mg2+和Mn2+对纳他霉素的生物合成无显著影响。【结论】不同种类、不同浓度的金属离子,对褐黄孢链霉菌菌体生长和纳他霉素生物合成的影响差别较大;在培养基中添加适当浓度和种类的金属离子,有助于提高纳他霉素的产量。
【Abstract】 【Objective】 The effects of several metal ions on the growth of Streptomyces gilvosporeus and biosynthesis of natamycin were investigated.【Method】 Shaking flask fermentation with single factor comparison experiment was adopted.【Result】 Results showed that Mg2+ and Ca2+ could promote the growth of S.gilvosporeus,Fe2+,Cu2+,Mn2+ and Co2+ inhibited the growth of S.gilvosporeus,while Na+,K+ and Zn2+ had no obvious effects on the growth of S.gilvosporeus.Adding appropriate concentrations of K+,Ca2+and Co2+ into production culture can stimulate the biosynthesis of natamycin,Na+,Fe2+,Cu2+ and Zn2+ inhibited the biosynthesis of natamycin,while Mg2+ and Mn2+ had no obvious effect on biosynthesis of natamycin.【Conclution】 Different metal ions at different concentrations could influence S.gilvosporeus cell growth and biosynthesis of natamycin at different degrees.Adding appropriate metal ions at proper concentrations can improve the production of natamycin by S.gilvosporeus.
