黄瓜细菌性角斑病菌的分离与鉴定

【摘要】 黄瓜细菌性角斑病是黄瓜生产中的重要病害之一,在陕西的危害严重,发病率在50%以上。本研究采用常规组织分离法和柯赫氏证病律从杨凌地区黄瓜细菌性角斑病标样中分离到1株病原菌,结合形态学、生理生化指标及16S rDNA序列分析,鉴定为丁香假单胞杆菌(Pseudomonas syringestrain)CP001。初步确定适合丁香假单胞杆菌CP001菌株生长的最佳pH为6~8,最适温度为22~29℃。

【Abstract】 Cucumber angular leaf spot is an important disease in cucumber production in Shaanxi.The incidence of cucumber caused by Pseudomonas syringae was over 50%.Through tissue plating and Koch’s pustule methods,one pathogen was isolated from diseased cucumber plant in Yangling of Shaanxi province.The pathogen was identified as Pseudomonas syringe strain CP001 based on morphological characteristics,cultural physiological and chemical characteristics,and the sequence of 16S ribosomal DNA.Biological characteristics of strain CP001 had been preliminarily studied.Results showed that optimal pH was 6-8,and temperature was 22-29 ℃.
