中国5个地区家养双峰驼的遗传多样性研究

【摘要】 为了科学评价、保护和利用骆驼遗传资源,采用10个微卫星位点对40峰阿拉善骆驼、40峰河西骆驼、60峰北疆骆驼、60峰东疆骆驼和60峰南疆骆驼共260个个体进行群体间、群体内遗传水平变化研究,并对群体结构进行遗传多样性分析。检测得到129个等位基因。5个群体的多态信息含量(PIC)为0.578~0.665,观察杂合度(Ho)的范围为0.187~0.261,期望杂合度的变化范围为0.612~0.699。依据Da遗传距离构建的UPGMA系统发生树表明,5个地区家养双峰驼可以分为两个分支,阿拉善骆驼、河西骆驼、东疆骆驼聚为一支;北疆骆驼和南疆骆驼聚为一支。结果表明,河西骆驼和阿拉善骆驼显示了最密切的关系,而阿拉善骆驼和南疆骆驼之间有最远的遗传距离。

【Abstract】 The domesticated bactrian camel in Chinese five areas(40 Alashan camel,40 Hexi camel,60 Beijiang camel,60 Dongjiang camel,and 60 Nanjiang camel) were analyzed the genetic variation within the population and the genetic relationship and structure among the populations by using 10 microsatellite markers.A total of 129 microsatellite alleles were detected.The UPGMA phylogenetic tree was constructed based on Da genetic distance and two clusters were obtained,Alashan camel,Hexi camel and Dongjiang camel formed one cluster,Beijiang camel and Nanjiang camel formed another cluster.The results suggested that the domesticated bactrian camel in Chinese five areas were separated two clusters by Structure2.2 software(K=2),and the population relationship is nearly the same to the UPGMA phylogenetic tree.Hexi camel had the closed relationship with Alashan camel which had the maximum genetic distance with Nanjiang camel.
