甲醛对雌性大鼠卵巢结构及卵母细胞超微结构和凋亡的影响

【摘要】 目的:研究甲醛对雌性大鼠卵巢病理组织结构、卵母细胞超微结构及卵母细胞凋亡的影响。方法:将40只SD雌性大鼠随机分为正常组和3个不同浓度甲醛染毒组,腹腔注射甲醛,剂量分别为20.0、2.0和0.2 mg/kg,每天1次,连续14 d,14 d后处死所有大鼠取卵巢组织,应用HE染色光镜观察雌性大鼠卵巢组织病理形态变化,应用电镜技术观察雌性大鼠卵母细胞超微结构的变化,应用TUNEL染色观察卵母细胞凋亡的情况。结果:甲醛可引起雌性大鼠卵巢组织的病理结构变化,其病理变化的程度与甲醛呈剂量依赖性;电镜检查发现甲醛可破坏卵母细胞的超微结构,其超微结构变化的程度与甲醛亦呈剂量依赖性;TUNEL染色观察到甲醛染毒组大鼠卵母细胞的凋亡率均高于对照组,且中、高剂量组和对照组间凋亡率比较差异具有显著性意义。结论:甲醛对雌性大鼠的卵巢组织结构及卵母细胞超微结构造成不良影响,促使卵母细胞凋亡。

【Abstract】 AIM: To study the effects of formaldehyde on ovarian histopathology structure,oocyte ultrastructure and apoptosis of female rat.METHODS: Healthy female rat were injected intraperitoneally with different doses of formaldehyde(20.0,2.0,0.2 mg/kg) continuously for 14 days.These mice were killed at the 14th day.Morphological changes of ovary cells were observed by light microscopy and ultrastructure of oocyte were observed by electron microscope.The oocyte apoptosis was determined by terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling(TUNEL) staining.RESULTS: By light microscopy,pathological structure changes in ovarian of female rat after treated with formaldehyde,and the changes in the rat ovarian increased with dose.By electron microscope,oocyte ultrastructure changes in ovarian of female rat after treated with formaldehyde too,and the changes in the mouse ovarian increased with dose.The results also showed that formaldehyde could cause the apoptosis in the female mouse ovarian compared with the control.And in middle and high doses,the difference between the apoptosis rate is distinctive.CONCLUSION: Formaldehyde has adverse effects on ovarian histopathology structure,oocyte ultrastructure of femal rats and it can cause the apoptosis of oocyte.
