PNGF复合材料的神经生长因子缓释性能

【摘要】 应用自行设计的RGD接枝的聚(羟基乙酸-L-赖氨酸-乳酸),复合聚乳酸、β-磷酸三钙和神经生长因子(NGF)制备神经生长因子缓释材料(PNGF)。在体外应用酶联免疫吸附检测法对NGF的释放量进行检测,并应用PC-12细胞培养法检测NGF活性。结果显示,NGF能够持续释放达到30 d,且保留80%以上的比活性,第26 d释放的NGF依然能够显著提高PC-12细胞在无血清培养液中的存活。

【Abstract】 Based on diffusion of NGF from degradable material,a novel nerve repair material with sustained NGF release was prepared by embedding NGF into a degradable RGD-modified composite.In vitro,the NGF release was quantified using enzyme-linked immunosorbent assay method,and the bioactivity of released NGF was examined by culturing PC-12 cells.The results showed that the NGF release process was prolonged over 30 days,and NGF bioactivity has been preserved after fabrication.The NGF released at the day 26th day could still significantly enhance the survival of PC-12 cells in serum-free medium.
