抗氧化剂对急性肺损伤大鼠细胞因子的影响研究

【摘要】 目的:探讨急性肺损伤(ALI)的发病机制及抗氧化剂对ALI大鼠细胞因子的影响。方法:健康、成熟Wistar大鼠60只随机平均分为模型组和治疗组,2组均以大肠杆菌腹腔注射,建立ALI动物模型。治疗组在注射大肠杆菌30min后,经大鼠尾静脉注射还原型谷胱甘肽。分别于注射大肠杆菌后3、6、12h测定2组血中IL-8、IL-10的变化。结果:模型组注射大肠杆菌后3h出现ALI症状,6h更为明显,12h口吐粉红色分泌物,解剖后肉眼所见双肺有明显水肿及出血点,氧合指数及肺湿/干重比均达到ALI诊断标准。治疗组3h无明显变化,6h呼吸稍促,12h肺组织仅轻度水肿。在使用还原型谷胱甘肽后,治疗组肺湿/干重比各时段检测值均低于模型组,其中6、12h差异有统计学意义(P<0.05);IL-8检测值各时段均低于模型组,差异均有统计学意义(P<0.05);IL-10检测值各时段均高于模型组,6、12h差异有统计学意义(P<0.05)。结论:ALI时存在炎性介质的失控与失衡,应用还原型谷胱甘肽后可有效调控炎性介质,从而降低ALI的发生概率。

【Abstract】 Objective:To investigate the pathogenesis of acute lung injury (ALI), and the effect of anti-oxidants on cytokines of ALI in rats. Methods:Sixty Wistar rats were randomly divided into 2 groups, the model group and the treatment group. The animal model of ALI was established by intraperitoneal injection of Escherichia(E.)coli. The anti-oxidant, reduced glutathione, was injected by the rat tail vein 30 min after injection of E. coli in rats of treatment group. The levels of serum IL-8 and IL-10 were detected 3, 6 and 12 h after injection of E. coli. Results:The symptom of ALI appeared 3 h after injection of E. coli, and was more serious after 6 h in rats of the model group. There were anatomical lung edema and bleeding points 12 h after injection of E. coli in rats of the model group. The oxygenation index and lung wet/dry weight ratio reached the diagnostic criteria of ALI. There was no significant change after 3 h, rapid breath after 6 h and only mild lung edema after 12 h in rats of the treatment group. The value of lung wet/dry weight ratio was lower in rats of treatment group than that of model group, which was significantly different after 6 and 12 h treatment (P < 0.05). The levels of IL-8 w ere lower at various time points in treatment group than those of model group(P < 0.05). The levels of IL-10 were significantly higher at different detection time points in treatment group than those of model group (P < 0.05). Conclusion:There was an imbalance and uncontrol of inflammatory mediators in acute lung injury. After application of reduced glutathione, inflammatory mediators can be effectively controlled, thereby reducing the occurrence probability of the acute lung injury.
