糖尿病肾病患者血清同型半胱氨酸与氧化应激的关系
The Relationship between the Serum Homocysteine and Oxidative Stress in Patients with Diabetic Nephropathy
【摘要】 目的:探讨血清高同型半胱氨酸(Hcy)导致的氧化应激与糖尿病肾病的关系。方法:将150例2型糖尿病(T2DM)患者按照尿蛋白水平分为正常蛋白尿组、微量蛋白尿组及临床蛋白尿组,各50例,随机抽取50例健康者为对照组。用酶联免疫吸附试验(ELISA)法检测血清Hcy,黄嘌呤氧化酶法测定血清中超氧化物歧化酶(SOD)、二硫代硝基苯甲酸比色法(DNTB法)测定还原型胱甘肽(GSH)、硫代巴比妥法(TAB法)测定丙二醛(MDA)的含量。结果:T2DM患者血清Hcy、MDA含量均高于对照组(P<0.01);微量蛋白尿组及临床蛋白尿组Hcy、MDA高于正常蛋白尿组(P<0.01),T2DM患者血清SOD、GSH含量低于对照组(P<0.01);微量蛋白尿组及临床蛋白尿组Hcy、MDA低于正常蛋白尿组(P<0.01);Hcy>15μmol/L组的血清SOD、GSH水平低于Hcy≤15μmol/L组(P<0.01),MDA水平高于Hcy≤15μmol/L组(P<0.01)。T2DM患者血清Hcy水平与血清中MDA水平呈正相关(r=0.64,P<0.01),与SOD、GSH含量均呈负相关(r分别为-0.57和-0.62,均P<0.01)。结论:血清Hcy导致氧化应激损伤参与糖尿病肾病的发生、发展过程。
【Abstract】 Objective:To investigate the relationship between the level of serum maleic dialdehyde(MDA),superoxide dismutase(SOD)and reduced glutathione(GSH)with complication of type 2 diabetic mellitus.Methods:One hundred and fifty patients with type 2 diabetes mellitus were divided into three groups,patients with normal albuminuria,microalbuminuria and clinical proteinuria group(n = 50 for each group),50 healthy persons were as control group.The level of serum homocysteine(Hcy) was detected by ELISA assay.The levels of serum SOD,GSH and MDA were detected by spectrophotometer.Results:The serum levels of Hcy and MDA were significantly higher in patients with diabetes than those of normal control group(P < 0.01),and significantly higher in micro-albuminuria albuminuria group and clinical group than those of normal albuminuria group(P < 0.01).The levels of SOD and GSH were significantly lower in patients with diabetes than those of normal control group(P < 0.01),and significantly lower in micro-albuminuria albuminuria group and clinical albuminuria group than those of normal control group(P < 0.01).The serum levels of SOD and GSH were significantly lower in Hcy>15 μmol/L group than those of Hcy≤15 μmol/L group(P < 0.01).The serum level of Hcy was positively correlated with the serum level of MDA(r = 0.64,P < 0.01) and negative correlated with SOD and GSH(r =-0.57 and r =-0.62,P < 0.01).Conclusion:The serum homocysteine involved in the development and procession of diabetic nephropathy via damaged oxidative stress.
