富锌中药驱铅丸对低浓度铅接触大鼠海马区NO及学习记忆能力的影响

【摘要】 目的:从海马区nNOS、iNOS表达及NO生成等方面探讨低浓度铅接触大鼠行为记忆的改变及富锌中药驱铅丸的干预机制。方法:40只大鼠随机分为5组:空白组饮双蒸水;其他4组饮水中均添加0.02%醋酸铅,其中高低剂量组分别按驱铅丸3.5g/kg.d-1和2.0g/kg.d-1的剂量2ml.d-1灌服;Ca-EDTA治疗组依地酸钠钙加普鲁卡因肌肉注射,50mg/kg.d-1,连续注射4天为1个疗程,休息4天后进行下一个疗程;连续治疗7个疗程;模型组不予任何治疗。60天后,进行Morris水迷宫试验,原子吸收光谱检测全血、脑组织铅含量;检测脑组织NO含量和NOS活性;RT-PCR检测脑组织海马区nNOS mRNA表达,West-ern blotting方法检测海马区nNOS蛋白表达。结果:与空白组比较,模型组血铅、脑铅含量显著增高(P<0.01),与模型组比较驱铅丸高剂量组脑铅、血铅含量显著降低(P<0.05);Morris水迷宫试验结果提示模型组大鼠空间定位航行能力下降(P<0.05),中药治疗组定位能力较模型组明显提高,具有统计学意义(P<0.001),模型组NO、NOS显著高于空白组(P<0.01);与模型组比较,各治疗组NO、NOS含量均显著降低(P<0.05);与空白组比较,模型组nNOS mRNA及蛋白表达显著降低(P<0.01),iNOS显著增高;与模型组比较,高剂量组nNOS mRNA及蛋白表达显著增高(P<0.05),iNOS表达显著降低(P<0.05)。结论:驱铅丸可以改善铅接触大鼠的行为记忆障碍,其机理与抑制iNOS激活,生成NO有关。

【Abstract】 Objective:To discuss the learning and memory changes of rats with low concentration lead and the mechanism of intervening effect of zinc rich Chinese medicinequqian pill on it from hippocampus nNOS and iNOS expression and NOformation.Methods:40 rats were divided into 5 groups,normal,model,positive,quqian pill low and high dose groups,model group was given 0.02%lead acetate orally,the low and high dose quqian pill groups were given Chinese medicine 3.5g/kg and 2.0g/kg respectively on the basis taking 0.02%lead acetate orally,positive group was given Ca-EDTA.Morris test was used to determine the learning and memory abilities of rats,examining the lead concentration of blood and brain,hippocampus nNOS mRNA was determined by RT-PCR,hippocampus nNOS protein was tested by Western blotting.Results:The lead concentration of blood and brain in model group was higher than that of normal group,quqian pill could significantly lower the lead content. Morris test showed that the learning and memory abilities of model rats were lowered,quqian pill could improve the abilities,there was difference between the model and Chinese groups.The hippocampus NO and NOS in model rats were higher than those in normal,which were lowered significantly in each treatment group.The hippocampus nNOS mRNA and protein expression were lowered and iNOS was raised in model group,the hippocampus nNOS mRNA and protein expression were elevated and iNOS expression was reduced obviously in the high dose group.
