消炎痛对外阴阴道假丝酵母菌病小鼠阴道组织中IL-10和IL-12p70的影响

【摘要】 目的研究消炎痛对外阴阴道假丝酵母菌病小鼠阴道组织中细胞因子IL-10和IL-12p70的影响,进一步探索消炎痛治疗外阴阴道假丝酵母菌病的可能机制。方法 37只昆明系小白鼠随机分成3组,雌激素诱导的外阴阴道假丝酵母菌病组(EI组,n=15),消炎痛治疗EI组(Indo组,n=15),空白对照组(C组,n=7)。分别在白假丝酵母菌接种后的第4、7、14天每组随机选取5只小鼠经阴道灌洗后处死,取阴道组织。用PAS染色进行阴道组织病理学观察,并用ELISA方法检测小鼠阴道组织中IL-10和IL-12p70水平。结果 PAS染色结果显示消炎痛可抑制白假丝酵母菌菌丝的生长。与C组相比,EI组阴道组织中的IL-10水平明显增高,但IL-12p70水平没有明显差异。与EI组相比,Indo组每个时点的IL-10水平明显下降,相反IL-12p70的水平明显增加。结论消炎痛不仅可抑制菌丝的生长,同时可通过降低阴道组织中IL-10的水平和增加阴道组织中IL-12p70的水平,影响阴道局部免疫力,从而抑制假丝酵母菌阴道炎的发生发展。

【Abstract】 Objective To investigate the effect of indomethacin on the expression of interleukin-10(IL-10)and interleukin-12p70(IL-12p70)in vaginal tissue of murine model of vulvovaginal candidiasis and further explore the possible mechanism of indomethacin in the treatment of vulvovaginal candidiasis.Methods Thirty-seven female Kunming mice were randomly divided into three groups:estrogen-induced vulvovaginal candidiasis group(EI group,n=15),indomethacin-treated group(Indo group,n=15)and control group(C group,n=7).The vaginal tissues were obtained at the day 4,7 and 14 after the inoculation.PAS dying was used to observe the pathological changes in vaginal tissues.Enzyme-linked immunosorbent assay(ELISA)was applied to determine the levels of IL-10 and IL-12p70 in the vaginal tissues.Results PAS dying showed that indomethacin could inhibit the hypha forming.As compared with C group,the levels of IL-10 in EI group were significantly increased,but the levels of IL-12p70 had no significant difference.As compared with EI group,the levels of IL-10 in Indo group were significantly decreased,but those of IL-12p70 were significantly increased.Conclusion Indomethacin can inhibit the development of vulvovaginal candidiasis not only by inhibiting the hypha forming,but also by affecting local immunity in vaginal tissues through down-regulating IL-10 and up-regulating IL-12p70 in vaginal tissues.
