【摘要】 目的研究神经颗粒素蛋白在系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)中的表达与调节。方法采用Western blot检测37例SLE患者PBMCs神经颗粒素蛋白的表达状态,同时用ELISA和放射免疫法分别测定其血浆IL-2和游离三碘甲状腺原氨酸(FT3)含量。结果 78.4%(29例)的SLE患者PBMCs表达神经颗粒素蛋白,而健康对照人群仅有29.4%的阳性表达率,其差异有统计学意义(连续性校正χ2=9.969,P=0.002)。且SLE患者神经颗粒素蛋白表达水平亦高于健康对照组,但差异无统计学意义(t=1.857,P>0.05)。神经颗粒素表达与血浆IL-2水平呈负相关(rs=-0.644,P<0.01),与血浆FT3水平无相关性(rs=-0.252,P=0.195)。结论神经颗粒素蛋白高表达于SLE患者PBMCs,炎症性细胞因子如IL-2可能参与其表达调节。更多还原

【Abstract】 Objective To observe the ectopic expression and regulation of neurogranin protein in peripheral blood mononuclear cells(PBMCs)of patients with systemic lupus erythematosus(SLE).Methods The neurogranin protein of PBMCs from 37 cases of SLE and 17 cases of healthy controls was detected by using Western blot.The levels of interleukin-2(IL-2)and free Triiodothyronine(FT3)in plasma were determined by ELISA and RIA,respectively.Results 78.4%(29 cases)SLE patients and 29.4%(5 cases)healthy controls showed a detectable neurogranin protein expressed in PBMCs.There was significant difference between two groups(continuity correction χ2=9.969,P=0.002).SLE patients had higher neurogranin protein level expressed in PBMCs(0.643 6±0.348 0)than in healthy controls(0.345 5±0.176 9),but no difference was found(t=1.857,P>0.05).The expression of neurogranin protein in SLE PBMCs was negatively correlated to IL-2 level in plasma(rs=-0.644,P<0.01),but not to FT3 level(rs=-0.252,P=0.195).Conclusion There was ectopic overexpression of neurogranin protein in PBMCs from SLE patients,perhaps resulting from the fluctuations of inflammatory cytokines such as IL-2.更多还原
