TGF-βRⅡ、NF-κB在肺癌中的表达及肿瘤相关性巨噬细胞计数与肺癌临床病理特征的关系

【摘要】 目的通过对肺腺癌和肺鳞状细胞癌中转化生长因子-βⅡ型受体(TGF-βRⅡ)、核转录因子-κB(NF-κB)的表达及肿瘤相关性巨噬细胞(TAMs)计数的研究,探讨三者与肺腺癌和肺鳞状细胞癌的关系。方法应用免疫组织化学技术检测TGF-βRⅡ蛋白、NF-κB蛋白及肿瘤相关性巨噬细胞在肺腺癌和肺鳞状细胞癌组织中的表达,并与癌旁正常肺组织中的表达进行比较。结果①TGF-βRⅡ蛋白的表达在肿瘤组织中明显降低(P<0.01),其表达下调与肺癌的组织学类型、淋巴结转移、临床分期相关(均P<0.05)。②NF-κB蛋白的表达在肿瘤组织中明显增强(P<0.01),其表达增强与肿瘤大小、淋巴结转移、临床分期相关(P<0.01,P<0.05)。③TAMs在肿瘤间质内浸润数量明显增多(P<0.01),并与肺癌的组织学类型、肿瘤大小、淋巴结转移、临床分期密切相关(P<0.05,P<0.01)。④TGF-βRⅡ蛋白与NF-κB蛋白的表达呈负相关(P<0.01),NF-κB蛋白的表达与TAMs的浸润数量呈正相关(P<0.01)。结论 TGF-βRⅡ蛋白表达水平降低可能促进肺癌的发生、浸润和转移,对肺腺癌发生或进展可能影响更大;NF-κB可能通过下调TGF-βRⅡ的表达、吸引肿瘤相关性巨噬细胞至肿瘤间质而促进肿瘤的发生、发展,且这种作用在非小细胞肺癌中具有较普遍的意义。

【Abstract】 Objective To investigate the expression of TGF-β RⅡ,NF-κB and the amount of tumor-associated macrophages(TAMs)in lung adenocarcinoma and lung squamous cell carcinoma and their correlation.Methods The expression of TGF-βRⅡ,NF-κB and TAMs was detected by immunohistochemistry in lung adenocarcinoma group,lung squamous cell carcinoma group and control group.Results ①As compared with the control group,the expression of TGF-βRⅡ was decreased in lung adenocarcinoma group and squamous cell carcinoma group(P<0.01),and it was correlated with histological type,lymph node metastasis and clinical stage(P<0.05);②As compared with the control group,the expression of NF-κB was increased in lung adenocarcinoma group and squamous cell carcinoma group(P<0.01),and it was correlated with tumor size,lymph node metastasis and clinical stage(P<0.01,P<0.05);③As compared with the control group,the number of TAMs was increased in tumor groups(P<0.01),and it was correlated with histological type,tumor size,lymph node metastasis and clinical stage(P<0.05,P<0.01);④The expression of TGF-βRⅡ was negatively correlated with that of NF-κB p65 protein(P<0.01).The number of TAMs was positively correlated with the expression of NF-κB p65 protein(P<0.01).Conclusion Low expression of TGF-βRⅡ may be related with the tumorigenesis of lung and carcinomatous growth,invasion and metastasis.This effect may be more significant in lung adenocarcinoma than in squamous cell carcinoma.NF-κB probably promotes tumorigenesis through down-regulating the expression of TGF-βRⅡ and attracting TAMs into tumor stroma.There is a more general significance of the effect in non-small cell lung cancer.
