基于毛细管电泳-安培检测的新生儿苯丙酮尿症早期无创诊断

【摘要】 首次采用毛细管电泳-安培检测法(CE-AD)同时测定了苯丙酮尿症(Phenylketonuria,PKU)患者尿液中的两种标志物:具有电活性的苯丙酮酸及非电活性的苯乙酸。考察了运行液添加剂、SDS浓度、工作电极电位等因素对分离和检测的影响。在优化实验条件下,两种标志物在15 m in内可实现基线分离。苯丙酮酸和苯乙酸的检出限(S/N=3)分别为0.50、0.92 mg/L。苯丙酮酸、苯乙酸浓度与峰高分别在3.0~1.0×103mg/L、2.0~5.0×103mg/L范围内呈良好线性,峰高与迁移时间的相对标准偏差分别为4.4%、3.2%(苯丙酮酸)和2.2%、4.3%(苯乙酸)。该方法已成功用于PKU患儿尿液中两种标志物的测定,且完全避免了尿液中尿酸对标志物测定的干扰,有望用于PKU患儿的早期无创诊断。

【Abstract】 A capillary electrophoresis with amperometric detection(CE-AD) method was developed for the simultaneous determination of the marked compounds of phenylketonuria(PKU),including electroactive phenylpyruvic acid and non-elecroactive phenylacetic acid,in infant urine samples.Several important parameters such as running buffer additive,sodium dodecyl sulfate(SDS) concentration and working electrode potential were investigated.Under the optimum conditions,phenylpyruvic acid and phenylacetic acid could be well separated in 15 min with 60 mmol/L borate running buffer(pH 7.8) containing 20 mmol/L SDS and 0.018 mmol/L 3,4-dihydroxybenzylamine(DHBA).The detection limits(S/N=3) were 0.50 mg/L and 0.92 mg/L,respectively.The concentrations and peak heights of phenylpyruvic acid and phenylacetic acid were in good linearities in the range of 3.0-1.0×103 mg/L and 2.0-5.0×103 mg/L,respectively.The RSDs of the peak heights and migration times for two marked compounds were 4.4%,3.2%(phenylpyruvic acid) and 2.2%,4.3%(phenylacetic acid),respectively.Being free from the interference of coexisting compound uric acid in urine samples,the proposed method was successfully applied in the simultaneous determination of two PKU marked compounds in infant urine samples after simple preparation,which could provide an alternative for the early noninvasive diagnosis of PKU infants.
