苦参碱对小鼠免疫器官的影响
Effect of Matrine on the Immunological Organs of Kunming Mice
【摘要】 目的观察苦参碱(MT)对昆明小鼠免疫器官的影响。方法将40只昆明小鼠分为苦参碱组、5-Fu组、苦参碱+5-Fu组和生理盐水(NS)阴性对照组。给药组小鼠分别按40,30,20+30mg/kg腹腔注射给药。计算不同组别小鼠的胸腺指数,脾脏指数和肝脏指数,并进行病理组织学观察。结果苦参碱组小鼠的胸腺、脾脏和肝脏指数均无显著变化(P>0.05);5-Fu组小鼠胸腺和脾脏指数均显著降低(P<0.01);苦参碱+5-Fu组小鼠的胸腺指数与对照组相比显著降低(P<0.01)。病理组织学光镜观察结果显示,苦参碱组小鼠胸腺和脾脏没有明显的萎缩和破坏,肝脏组织无明显病理变化。结论苦参碱可以拮抗5-Fu对脾脏的免疫抑制,使脾脏指数增加。
【Abstract】 Objective To investigate the effect of Matrine on the immunological organs of Kunming mice. Methods 40 Kunming mice were divided randomly into 4 groups: Matrine treated,5-Fu treated,Matrine + 5-Fu treated and control groups,subjected to 40,30,20 + 30 mg/kg and normal saline through abdominal cavity injection,respectively. The weight and index of thymus gland,spleen and liver of mice were measured. The tissues of thymus,spleen and liver were observed by pathological examination. Results Matrine had no significant effect on the weight and indexes of thymus,spleen and liver in Kunming mice( P > 0. 05) ; 5-Fu decreased the weight and indexes of thymus and spleen( P < 0. 01) ; Matrine + 5-Fu decreased the weight and index of thymus ( P < 0. 01) . The weight and index of spleen matrine + 5-Fu treated group were higher than those of 5-Fu treated group ( P < 0. 01) while almost the same as those of control and Matrine treated group( P > 0. 05) . H&E staining showed the structure of thymus,spleen and liver was almost normal in Matrine-treated group. Conclusion Matrine can antagonize immune suppression induced by 5-Fu and raise the weight of spleen in mice.
