细胞周期相关激酶在卵巢癌细胞中的亚细胞定位及其表达的意义
Subcellular localization of cell cycle-related kinase in ovarian carcinoma cells and significance of its overexpression in ovarian carcinoma tissues
【摘要】 目的分析细胞周期相关激酶(CCRK)在卵巢癌细胞中的亚细胞定位,研究CCRK在正常卵巢组织与卵巢癌组织中的表达差异,分析CCRK表达与卵巢癌患者预后的关系。方法采用激光共聚焦显微镜确定CCRK在卵巢癌细胞中的亚细胞定位,应用免疫组织化学染色(IHC)检测130例卵巢癌组织与30例正常卵巢组织中CCRK表达水平,并将CCRK表达水平与患者预后进行分析。结果激光共聚焦显微镜检测表明CCRK在卵巢癌细胞中的表达主要定位于细胞质和核膜周围,细胞核内也有少量分布。与正常卵巢组织相比,卵巢肿瘤组织中CCRK表达水平明显升高(P<0.05),CCRK表达上调与卵巢癌患者预后呈负相关(P<0.001)。结论 CCRK在卵巢癌细胞中的表达主要定位于细胞质和核膜周围,其在卵巢癌组织中表达异常上调与患者预后密切相关,提示CCRK在卵巢癌发生、发展过程中可能起重要作用。
【Abstract】 Objective To determine the subcellular localization of cell cycle-related kinase(CCRK) in ovarian carcinoma cells,compare CCRK expression levels between ovarian normal and carcinoma tissues,and investigate the relationship between CCRK overexpression and survival in patients with ovarian carcinoma.Methods Confocal microscope was employed to determine CCRK subcellular localization in ovarian carcinoma cells,immunohistochemistry(IHC) staining was used to detect CCRK expression in 130 ovarian carcinoma tissues and 30 normal tissues,and the relationship between CCRK overexpression and survival of patients with ovarian carcinoma was further analyzed.Results Determined by confocal microscope,it was found that CCRK expressed mainly in the cytoplasm and peri-nuclear membrane area with minor expression in the nucleus;compared with ovarian normal tissues,CCRK expression was upregulated in ovarian carcinoma tissues(P<0.05);CCRK overexpression was negatively related to survival of ovarian carcinoma patients(P<0.001).Conclusions CCRK expresses mainly in the cytoplasm and peri-nuclear membrane area;compared with ovarian normal tissues,CCRK expression is upregulated in ovarian carcinoma tissues and negatively related to survival of patients with ovarian carcinoma,which possibly suggests a pivotal role of CCRK in the carcinogenesis and development of ovarian carcinoma.
