凋亡相关基因PDCD5表达与SLE临床相关性的研究

【摘要】 目的:通过检测程序化细胞死亡因子5(programmed cell death5,PDCD5)在系统性红斑狼疮(systemic lupus erythematosus,SLE)中的表达,探讨其与SLE发生与发展、自身抗体产生和肾脏损伤的关系。方法:采用RT-PCR和Western blot法,分别从mRNA和蛋白质水平检测PDCD5在67例SLE患者和50例健康对照外周血单个核细胞(PBMC)中的表达。结果:SLE活动期患者PDCD5的阳性表达率和表达水平显著高于健康对照组(P<0.01);活动期患者PDCD5的表达水平显著高于稳定期患者(P<0.01),阳性表达率相比差异无显著性;SLE稳定期患者PDCD5的表达量显著高于健康对照组(P<0.01),但阳性表达率差异也无显著性。有2种或2种以上自身抗体阳性的SLE患者PDCD5表达量显著高于有1种或无自身抗体阳性的SLE患者(P<0.01);SLE活动期合并狼疮性肾炎患者和无合并狼疮性肾炎患者的PDCD5表达差异无显著性(P>0.05)。结论:SLE患者PBMC中PDCD5的表达上调,活动期患者升高更为明显,提示PDCD5可能通过增加表达量而导致异常的淋巴细胞凋亡参与SLE发生和发展。

【Abstract】 Objective To explore the association of programmed cell death 5(PDCD5) gene with the genesis and development of systemic lupus erythematosus(SLE), autoantibody production, and renal damage by detecting PDCD5 expression in SLE.Methods The expressions of PDCD5 mRNA and protein in PBMC from 67 SLE patients and 50 healthy controls were detected by semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR) and Western blot.Results The positive rate and expression level of PDCD5 were significantly higher in active SLE patients than in normal controls(P < 0.01).The expression level of PDCD5 was significantly higher in active SLE than in inactive SLE(P < 0.01), but PDCD5 positive rate did not differ between active and inactive SLE patients.The expression level of PDCD5 was significantly higher in inactive SLE patients than in normal controls(P < 0.01), while there was no significant difference in PDCD5 positive rate between inactive SLE patients and normal controls.The expression level of PDCD5 was significantly higher in patients with at least two types of autoantibodies than in those with only one type of autoantibody or none(P < 0.01).No significant difference of PDCD5 expression was found between active SLE patients with lupus nephritis and those without lupus nephritis(P > 0.05).Conclusions The up-regulation of expression of PDCD5 in SLE patients, especially in those with active SLE, suggests that abnormal apoptosis of lymphocytes due to the overexpression of PDCD5 plays an important role in the pathogenesis of SLE.
