卡氏肺孢子虫肺炎大鼠模型的建立

【摘要】 目的采用SD大鼠建立卡氏肺孢子虫肺炎(Pneumocystis Carinii pneumonia,PCP)动物模型。方法 20只SD大鼠随机分成实验组和对照组。实验组皮下注射地塞米松2.5 mg/只,每周2次,对照组不给药。8周后,分别取肺组织,印片,Giemsa染色检查卡氏肺孢子虫滋养体和包囊。酚氯仿法提取大鼠肺组织DNA,LAMP法扩增卡氏肺孢子虫DNA。结果诱导后24 d大鼠出现死亡,28 d检获PC,32 d感染阳性率为86%(13/15);LAMP扩增出卡氏肺孢子虫特异DNA。结论成功诱导SD大鼠的PCP模型。

【Abstract】 Objective To establish the animal models of Pneumocystis Carinii pneumonia(PCP) with SD rats.Methods Twenty SD rats were randomly divided into experimental group and control group.The rats in experimental group were injected subcutaneously with Dexamethasone twice every week,while the rats in control group were injected with nothing.8 weeks later,the PC trophozoites or cysts in the lung print smears were observed by Giemsa staining.Phenol chloroform method was used to extract the DNA of lung tissues,and the LAMP method was used to amplify the DNA of PC.Results Twenty-four days later,death appeared in the rats of experimental group and PC was detected on the 28th day.On the 32nd day,the detectable rate of PC trophozoites or cysts in experimental group was 86%(13/15).The DNA of PC was detected in the lung tissues of the rats of experimental group.Conclusion The animal models of PCP are established by hypodermic injection of Dexamethasone in SD rats.
