【摘要】 目的观察EB病毒(EBV)潜伏膜蛋白1(LMP-1)在鼻咽癌(NPC)鼻咽拭子中的表达,探讨LMP-1表达与NPC临床病理特征的关系。方法纳入2007年～2008年福建省肿瘤医院放疗科收治的经病理检查确诊为NPC且同意参加该试验的初治患者129例,均在治疗前应用鼻咽拭子法取鼻咽部黏膜脱落细胞,提取DNA,采用PCR法检测LMP-1基因的表达。结果 129例患者中有114例患者LMP-1呈阳性表达,另15例呈阴性表达,阳性率为88.37%(114/129);对照组中无1例呈阳性表达。LMP-1阳性表达率有颈部淋巴结转移组明显高于无颈部淋巴结转移组。LMP-1表达与患者年龄、性别、临床分期、T分期及M分期无相关性。结论 PCR法检测鼻咽拭子中LMP-1的检出率高于免疫组化法,LMP-1表达与鼻咽癌患者颈部淋巴结转移密切相关,临床中可预测鼻咽癌患者的转移潜能,LMP-1可能做为抗肿瘤转移靶向治疗的潜在靶点。更多还原

【Abstract】 Objective To investigate the clinical significance of Epstein-Barr virus(EBV) latent membrane protein-1(LMP-1) in nasopharyngeal carcinoma(NPC).Methods 129 patients with pathologically confirmed NPC between 2007 and 2008 were involved in this study.After informed consent,nasopharyngeal swab was performed to obtain cells for the LMP-1 PCR assay.Results Of the 129 patients,LMP-1 was expressed in 114(88.37%).The positive expression rate of LMP-1 in patients with neck lymph node metastasis was significantly higher than that without neck lymph node metastasis(P < 0.05).The expression of LMP-1 was not correlated with age,gender,clinical stage,T stage,M stage and histological type(P > 0.05).Conclusion The detection rate of positive LMP-1 expression was higher by PCR assay than that by immunohistochemisty.The positive expression of LMP-1 was highly correlated with lymph node metastasis.It could be used to predict the metastatic potential of NPC,and further be used as a potential target for anti-metastasis therapy.更多还原
