肿瘤坏死因子-α-1031T/C基因多态性与甲状腺相关性眼病的相关性
Association of TNF-α gene-1031T/C polymorphism with thyroid-associated ophthalmopathy
【摘要】 目的探讨肿瘤坏死因子-α(TNF-α)-1031位点基因多态性与山西地区汉族人甲状腺相关性眼病(TAO)发病易感性的关系。方法研究对象51例TAO患者,56例Graves病(GD)患者和57例正常对照者,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法,检测研究对象TNF-α-1031基因型和等位基因的频率。结果①C等位基因TAO组频率明显高于GD组及正常对照组(29.4%vs13.4%,11.4%,P<0.05);TC+CC基因型频率TAO患者组明显高于GD患者组及正常对照组(54.9%vs26.8%,22.8%,P<0.05)。②按性别分层后,男性C等位基因频率TAO组高于GD组和正常对照组(26.1%vs9.1%,8.9%,P<0.05);TC+CC基因型频率TAO组与GD组及正常对照组比较均有显著性差异(47.8%vs18.2%,17.9%,P<0.05)。女性C等位基因频率TAO组高于正常对照组(30.4%vs13.8%,P<0.05);与GD组比,差异无统计学意义(P>0.05);TC+CC基因型频率TAO组与GD组及正常对照组比较均有显著性差异(60.7%vs32.4%,27.6%,P<0.05)。结论TNF-α-1031T/C基因多态性与山西地区汉族人TAO患者尤其是男性的遗传易感性有关。
【Abstract】 Objective To investigate the association of tumour necrosis factor-α(TNF-α)gene polymorphisms at position-1031 in Shanxi region of China.Methods The genotype and allele frequencies of TNF-α gene at position-1031 were measured in 51patients with TAO(TAO group),56 patients with Graves disease(GD group)and 57 normal controls(control group)by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method.Results ①The C allele frequencies and TC+CC genotype in TAO group were significantly higher than those in GD group or control group(P<0.05).②In the males,C allele frequency and TC+CC geno type frequencies in TAO group were higher than in GD group and control group(P<0.05).③In the females,C allele frequency in TAO group was higher than that in control group(P<0.05).But there was no significant difference between GD group and TAO group(P>0.05).TC+CC genotype frequency showed a significant difference from GD group and control group(P<0.05).Conclusion TNF-α gene-1031C allele may contribute to the development of TAO in Shanxi region of China,especially in male patients.
