鼻咽癌人源抗独特型单链抗体诱导的免疫应答
Immune Response Induced by Human Anti-idiotypic Antibody G22 ScFv against Nasopharyngeal Cancer
【摘要】 目的观察鼻咽癌人源抗独特型单链抗体G22ScFv在小鼠体内诱导的免疫应答,探讨其作为鼻咽癌疫苗的可能性。方法在大肠杆菌中诱导表达G22ScFv并进行纯化,以纯化的G22ScFv免疫BALB/c小鼠,采用间接ELISA及竞争抑制ELISA检测免疫小鼠特异性体液免疫应答水平;流式细胞术检测免疫小鼠脾T淋巴细胞表型的变化。结果纯化的融合蛋白纯度达95%以上,且具有良好的反应原性。经ELISA检测,G22ScFv免疫后,小鼠产生了Ab3,且小鼠脾脏CD4+、CD8+T淋巴细胞数量升高。结论G22ScFv可诱导机体产生体液免疫和细胞免疫应答,为鼻咽癌人源抗独特型抗体疫苗的临床应用奠定了基础。
【Abstract】 Objective To observe the immune response induced by human anti-idiotypic antibody G22 ScFv against nasopharyngeal cancer in mice and investigate the feasibility of G22 ScFv as a nasopharyngeal cancer vaccine.Methods G22 ScFv was expressed in E.coli,then purified and used for immunization of BALB /c mice.The specific humoral immune responses of mice were determined by indirect ELISA and competitive inhibition ELISA,and the splenic T lymphocyte phenotypes by flow cytometry.Results The expressed fusion protein reached a purity of more than 95% after purification and showed good reactogenicity.ELISA proved that G22 ScFv induced Ab3 and increased the counts of CD4+ and CD8+ T lymphocytes in spleens of mice.Conclusion G22 ScFv induced both humoral and cellular immune responses,which laid a foundation of clinical application of human anti-idiotypic antibody as vaccine against nasopharyngeal cancer.
