人参皂甙Rg1对人慢性粒细胞白血病p210 bcr/abl融合蛋白表达的影响

【摘要】 目的:研究人参皂甙Rg1对人慢性髓细胞白血病敏感株K562细胞p210bcr/abl融合蛋白表达的影响。方法:体外培养K562细胞,经人参皂甙Rg1处理不同时间后,用流式细胞仪检测细胞凋亡;用Western印迹技术检测细胞内p210bcr/abl融合蛋白表达。结果:人参皂甙Rg1可诱导细胞凋亡,凋亡率并随时间延长而升高;人参皂甙Rg1可下调p210bcr/abl蛋白表达量,并具时间依赖性。结论:人参皂甙Rg1可通过降低p210bcr/abl蛋白水平来诱导K562细胞凋亡,达到有效抗人慢性髓细胞白血病的效果。

【Abstract】 Objective:To investigate the effects of ginsenoside Rg1 on the expressions of p210 bcr/abl fusion protein in human chronic myeloid leukemia(CML).Methods:Human Ph+ CML K562 cells were cultured in vitro and treated with ginsenoside Rg1 for different time.Flow cytometry(FCM) was used to evaluate the apoptosis of leukemic cells.The protein levels of p210 bcr/abl in the leukemic cells were determined by Western blotting.Results:Flow cytometry showed that ginsenoside Rg1 can induce the apoptosis of K562 cells,and the percentage of apoptotic cells increased with treating time extending.Western blotting showed that ginsenoside Rg1 down-regulated the expressions of p210 bcr/abl fusion protein in a time-dependent manner.Conclusion:Ginsenoside Rg1may induce the apoptosis of K562 cells to inhibit CML effectively through down-regulating the expressions of p210 bcr/abl fusion protein.
