生物可降解塑料聚β-羟基丁酸酯产生菌的选育
Breeding of Producing Strains of the Biodegradable Plastic——Poly-β-hydroxybutyrate
【摘要】 作为一类可望替代传统塑料的新型可降解生物高分子材料,聚β-羟基丁酸酯(PHB)日益引起人们的重视。采用尼罗蓝荧光法结合苏丹黑染色法从污水筛选出产PHB的细菌,并发酵复筛得到一株PHB产量较高的菌株,PHB含量占细胞干重的16.64%,PHB产量为0.57 g/L。对其进行形态培养特征和生理生化特性研究,初步鉴定为动胶菌属,编号为Z-13。以Z-13为出发菌株进行紫外诱变,得到突变菌株uZ-13,PHB含量(21.54%)和PHB产量(0.78 g/L)分别比诱变前提高了29.45%和36.84%,且具有较好的遗传稳定性,为微生物法生产生物可降解塑料奠定基础。
【Abstract】 As a new kind of polymer material expected to replace the traditional plastics,poly-β-hydroxybutyrate(PHB) was paid great attention in recent years.Strains producing PHB were screened from the sludge by Nile blue fluorescence combined with the sudan black stain.A strain with higher PHB content was got by secondary screening with 16.64 % PHB content(account for the cell dry weight) and 0.57 g/L PHB yield.Colony characteristics,physiological and biochemical tests showed that the strain could be classified as Zoogloea Itzigohn and then the stain was named as Z-13.Mutation of the Z-13 with UV was studied.After mutagenesis,mutunt uZ-13 was got with PHB content(21.54 %) and yield(0.78 g/L) which respectively increased about 29.45 % and 36.84 %.Further study showed that uZ-13 possesses good genetic stability and laid a foundation for producing biodegradable plastics by microbiological method.
