新型防腐剂富马酸乙二醇甲酯的合成及抑菌活性研究

【摘要】 采用一锅法合成了具有α,β-不饱和羰基结构的新型防腐剂富马酸乙二醇甲酯。以摩尔比为1∶0.9的马来酸酐和乙二醇为原料,磷钨酸为催化剂,在140℃反应3 h得醇解产物马来酸单乙二醇酯;然后加入与马来酸酐摩尔比为2∶1的甲醇,回流酯化2 h,得到马来酸乙二醇甲酯;最后加入异构化催化剂浓盐酸,70℃下反应1 h,得到最终产品富马酸乙二醇甲酯,总收率达50.1%。抑菌活性实验表明:富马酸乙二醇甲酯对混合菌群的生长具有良好的抑制作用,其抑菌能力优于富马酸单甲酯,与苯甲酸相当。

【Abstract】 A new preservative glycol methyl fumarate(GMF) containing α,β-unsaturated carbonyl group was synthesized by one-pot reaction.Maleic anhydride and glycol(mole ratio 1∶0.9) reacted at 140 ℃ for 3h with phosphotunqstic acid as catalyst and monoglycol maleate was synthesized.Followed the esterification reaction between alcoholysis product and methanol.Mole ratio of methanol to maleic anhydride was 2∶1.This reaction was carried out under reflux for 2 h and the product was glycol methyl maleate.At last,the isomerization took place under the role of concentrated hydrochloric acid at 70 ℃.The isomerization reaction time was 1 h.The final product glycol methyl fumarate was obtained with the total yield up to 50.1%.The experiment results of antimicrobial activity indicate that GMF has good inhibiting activity for hybrid fungus.Its antimicrobial activity is better than mono-methyl fumarate and comparable to that of benzoic acid.
