微波预处理-热水浸提提取香椿叶多糖的工艺研究

【摘要】 研究微波预处理,热水浸提提取香椿叶总多糖的最佳工艺条件。以香椿叶总多糖提取量为考察指标,单因素考察了润湿时间、润湿料液比、微波功率、微波处理时间对香椿叶总多糖提取效果的影响。在单因素实验的基础上,用正交实验法对香椿叶总多糖的微波预处理工艺条件进行优化,选用L9(34)进行正交实验,通过实验设计优选出香椿叶总多糖微波预处理溶剂回流提取的最佳工艺条件并验证、确定提取次数。结果表明:在香椿叶质量10.0g,提取温度100℃,提取时间2h,浸提料液比1:15条件下,微波辅助处理香椿叶的最优工艺条件为:润湿料液比1:5,润湿时间1.5h,微波处理功率1000W,微波处理时间60s。提取次数5次。在最佳参数组合下,提取5次每克香椿叶可提取总多糖为46.156mg。

【Abstract】 The hot water extraction process of polysaccharide from Toona sinensis (A.Juss.) Roem.leaves assistend by microwave pretreatment was studied.The effect of the operation parameters such as microwave power,microwave pretreatment time,wetting time and the ratio of solid to wetting liquid on polysaccharides mass extracted from Toona sinensis (A.Juss.) Roem.leaves was analyzed with monofactor experiment.the optimum extraction process was optimized with orthogonal test L9(34) and verified,determined extracting times.The results showed that:on the condition that 10.0 g Toona sinensis (A.Juss.) Roem.leaves as the crude drug,extraction temperature 100 ℃,extraction time 2 h,the ratio of solid to extracting liquid 1:15 (g/mL),mass:volume),optimum microwave pretreatment conditions were determined as follows:the ratio of solid to wetting liquid 1:5 (g/mL,mass:volume),wetting time 1.5 h,microwave power 1000 W,microwave pretreating time 60 s.extracting times 5.The extracting mass of total polysaccharides from Toona sinensis (A.Juss.Roem.leaves was 46.156 mg/g extracted 5 times at the optimum parameters.
