海星多糖的分离纯化及鉴定
Isolation,purification and identification of starfish polysaccharide
【摘要】 目的:建立海星多糖分离纯化的方法,并对纯化组分进行鉴定。方法:用碱提-酶解法提取海星多糖,再用离子交换层析和凝胶过滤层析法进行纯化;用理化方法和光谱法对纯化组分进行鉴定。结果:分离纯化的海星多糖不含蛋白质,含硫酸基,其中SSP1含己糖醛酸18.5%,含氨基己糖22.6%,含岩藻糖16.8%。结论:获得海星多糖6个纯化组分(SSP1-6),其中SSP1由己糖醛酸、氨基己糖、岩藻糖和硫酸基组成,其组成摩尔比约为:1.00∶1.10∶1.07∶(未测含量)。
【Abstract】 Objective:The isolation and purification method of Starfish polysaccharide was set up and purified components was identified. Methods:Starfish polysaccharide was extracted by Alkaline extraction-enzymatic method and purified by ion-exchange chromatography and Gel-filtration chromatography. Purified components were identified using physical and chemical method and spectroscopy. Results:Purified starfish polysaccharide is free of protein,including sulfuric acid-bases. SSP1 contains 18.5% uronic acid,22.6% amino hexose,16.8% fucose. Conclusion:Six purified components (SSP1-6)were obtained and SSP1 is composed of uronic acid,amino hexose,fucose,sulfuric acid-bases with molar ratio of 1.00∶1.10∶1.07∶(not measured).
