【摘要】 目的探讨姜黄素对人口腔鳞癌细胞Tca8113及Tb增殖的抑制及诱导凋亡作用。方法分别应用6.25、12.50、25.00、50.00μmol/L的姜黄素作用Tca8113及Tb细胞24 h,采用MTT法检测姜黄素对口腔鳞癌细胞Tca8113及Tb增殖的抑制作用,形态学观察及流式细胞仪测定细胞凋亡。结果6.25~50.00μmol/L姜黄素能显著抑制口腔鳞癌细胞Tca8113和Tb细胞的增殖(F=793.94、391.08,q=6.00~62.68,P<0.05)。流式细胞仪检测显示口腔鳞癌细胞凋亡率随姜黄素浓度升高而增加。倒置显微镜下可观察到部分凋亡细胞出现染色质凝集、核固缩、核碎裂等形态学改变。结论姜黄素对人口腔鳞癌细胞Tca8113及Tb的增殖具有显著的抑制作用,并可诱导细胞凋亡。更多还原

【Abstract】 Objective To investigate the inhibition and apoptosis-inducing effects of curcumin on Tca8113 and Tb cells of human oral squamous cell carcinoma.Methods Tca8113 and Tb cells were treated with different concentrations of curcumin(6.25,12.50,25.00,and 50.00 μmol/L),respectively,for 24 hours.MTT was used to determine the inhibition of curcumin.Morphological assay and flow cytometry were employed to detect the apoptosis.Results The proliferation of Tca8113 and Tb cells were significantly inhibited by curcumin at all concentations applied(F=793.94,391.08;q=6.00-62.68;P<0.05).The results of flow cytometry demonstrated the apoptosis rate increased gradually along with the increase of concentrations of curcumin.Morphological changes including chromatic agglutination,karyopyknosis,and nuclear fragmentation could be observed in some of the cells under inverted microscope.Conclusion Curcumin can notably inhibit proliferation and induce apoptosis of Tca8113 and Tb cells of human oral squamous cell carcinoma.更多还原
