金堆城钼矿尾矿堆积区土壤细菌多样性分析

【摘要】 金堆城钼矿是我国目前最大的钼生产基地,钼矿的开采导致大量含有金属硫化物的尾矿堆积,使土壤及地下水环境发生了改变.为了解钼矿尾矿堆积后土壤微生物多样性的变化规律,分别采集5个不同时期堆积的尾矿土壤样品(S1~S5),应用16S rDNA V3区片段的变性梯度凝胶电泳(DGGE)技术分析土壤细菌群落结构的变化.结果显示S1~S5的DGGE条带图谱差异很大,且各有其特征条带.堆积时间较短的S1、S2的多样性明显大于堆积时间长的S3、S4、S5.对DGGE图谱中优势条带序列进行分析,土壤样品中的优势细菌主要属于变形菌门(Proteobacterium)、厚壁菌门(Firmicutes)、硝化螺旋菌门(Nitrospira)和一个未命名的菌种(Unidentified).本实验证实钼尾矿的堆积导致土壤细菌结构以及多样性发生改变,且多样性随着尾矿堆积时间的增加而减少;鉴定的优势菌种在微生物浸矿技术的发展和应用方面具有潜在的研究价值.

【Abstract】 Jinduicheng Molybdenum(Mo) Mine is the largest production base of Mo in China.A large number of tailings containing metal sulfides were accumulated as a result of exploitation of Mo mine,which made changes of the surrounding soil and groundwater environments.To investigate the variations of soil microbial diversity in response to the changes after Mo tailings accumulation,soil samples at different accumulated period(S1~S5) were collected at Mo Tailings areas.16S rDNA-V3-fragment based denaturing gradient gel electrophoresis(DGGE) was utilized to explore the variations of the soil bacterial structures.The results showed that DGGE patterns in different samples had displayed great variation,and each sample had its own characteristic strips.The diversities of samples S1 and S2 accumulated at short periods were significantly greater than the samples S3,S4 and S5 accumulated at relatively long periods.The sequence BLAST of the representative DGGE bands demonstrated that the preponderant bacteria in the studied soil samples mainly belonged to Proteobacterium,Firmicutes,Nitrospira and one unnamed strain(Unidetified).Based on the findings of this study,Mo tailings accumulation had caused changes in structure and diversity of the soil bacterial community.And the diversities of soil samples decreased along with the increasing accumulated time.The dominant bacteria isolated suggested that the microorganisms in Mo tailings had the potential research value in the development and application of microbial leaching of metallic elements.
