【摘要】 目的通过对db/db小鼠生殖功能障碍的基础研究,探讨中医"痰湿型"不孕症的病因病机——"痰瘀胞宫"的现代生物学基础,证明其理论的正确性。方法①雌性B6.Cg-m+/+Leprdb小鼠和其同窝出生的雌性野生型C57BL/6J小鼠于12周龄进行卵巢交互移植,构建实验小鼠的LR基因型组合分别为A组(躯体LR+,卵巢LR-,n=10)、B组(躯体LR+,卵巢LR+,n=5)、C组(躯体LR-,卵巢LR+,n=5)、D组(躯体LR-,卵巢LR-,n=5)共4组。②术后恢复两周,进行阴道脱落细胞涂片,连续观察两个周期。③取材,检测全身糖脂代谢指标和生殖激素水平,HE染色法检测卵巢、子宫组织形态学变化。结果①C、D组与A、B组比较,其体重增加约1倍(P<0.01),性腺周围脂肪垫重量增加约10倍(P<0.01),血清葡萄糖、低密度脂蛋白胆固醇(LDL-C)和胰岛素水平增加2～3倍(P<0.01)。②阴道细胞涂片发现,A、B组小鼠均出现正常的动情周期,而C、D组则始终处于动情间期;C、D组与A、B组比较,卵巢重量及E2、P、FSH、LH水平显著下降(P<0.05)。HE染色可见A、B组小鼠的卵巢及子宫组织形态正常,而C、D组小鼠的卵巢及子宫均萎缩变小。结论 db小鼠生殖功能障碍主要是高糖脂血症累及生殖器官导致其脂质化所引起,"痰湿型"不孕症的中医病因病机为"痰瘀胞宫",其生物学基础是糖脂代谢异常。更多还原

【Abstract】 Objective To explore the etiology and pathogenesis of infertility of phlegm damp type-the modern biological basis of "phlegm stasis in the uterus" in Chinese medicine by the basic study on db/db mouse reproductive dysfunction so as to prove the correction of this theory.Methods (1)Reciprocal transplantation of ovary was performed between female B6.Cg-m+/+ Lepr db mice and female wide type C57BL/6J mice in the same litter at the age of 12 weeks.And 4 LR genotype groups of the experimental mice were constructed,named A group(body LR+ and ovary LR-,n=10),B group((body LR+ and ovary LR+,n=5),C group(body LR-and ovary LR+,n=5)and D group(body LR-and ovary LR-,n=5).(2)2 weeks after postoperative recovery,vaginal smear was performed for continuous observations of 2 periods.(3)The samples were collected for the determination of general glucose and lipid metabolic indicators and reproductive hormone level.HE staining was adopted to determine the histomorphological changes in ovary and uterus.Results (1)As compared with A and B groups,the weight in C and D groups increased by around 1 time(P<0.01),the fat pad weight around gonad increased by about 10 times(P<0.01).The levels of blood glucose,LDL-L and insulin increased by 2 to 3 times(P<0.01).(2)It was discovered in vaginal smear that the normal estrous cycle happened in mice in A and B groups,but the mice in C and D groups were still in estrous period.Compared with A and B groups,the ovary weight in C and B groups as well as the levels of E2,P,FSH and LH decreased significantly(P<0.05).HE staining found that the ovary and uterus of mice in A and B groups were normal in histomorphology and those in C and D groups were getting smaller.Conclusion The db/db mice reproductive dysfunction is due to lipid-based reproductive organs induced by hyperglycemia and hyperlipidemia.In the etiology and pathogenesis of Chinese medicine,infertility of phlegm damp type is caused by "phlegm stasis in the uterus" and the abnormal glucose and lipid metabolism are the biological basis.更多还原
